摘要
为探讨油菜、蚕豆秸秆还田对稻田土壤氨氧化微生物群落结构的影响,以江汉平原碱性潮土为研究对象,设置单施化肥(NS)、油菜秸秆+单施化肥(RS)、蚕豆秸秆+单施化肥(BS)3个处理,借助Illumina Miseq高通量测序平台,探究不同类型秸秆还田对水稻土氨氧化古菌(AOA)和氨氧化细菌(AOB)群落多样性及群落结构的影响。结果表明,不同类型秸秆还田对AOB群落Alpha多样性指数的影响强于AOA。对于AOB群落,相比NS处理,秸秆还田(RS和BS)处理提高了其多样性指数。其中,RS处理的多样性指数高于BS处理。Veen分析结果显示,相比AOA,不同类型秸秆还田对AOB amoA基因序列OTU数目和分布的影响较大。其中,RS处理对OTU分布的影响强于BS处理。纲水平上,不同类型秸秆还田对AOA群落的物种组成及物种相对丰度没有显著影响(p>0.05)。但对于AOB群落,RS处理中unclassified_p__Proteobacteria的相对丰度比NS处理高2倍(p<0.05),比BS处理高1.45倍(p>0.05);相比NS处理,RS处理减少了norank_p__ammonia-oxidizing_bactera_ensemble的相对丰度,而BS处理缺失这一物种。综上可知,碱性潮土中AOB对不同类型秸秆还田的响应强于AOA。相比BS处理,RS处理对AOB群落结构的影响更大。
The purpose of this study was to investigate the effects of rape and broad bean straw returning on microbial community structure of ammonia oxidizers in paddy soil.Based on the alkaline soil of Jianghan Plain as the research object,the Illumina Miseq high-throughput sequencing platform was used to study the effects of different types of straw returning on the community diversity and structure of ammonia oxidizing archaea(AOA)and the ammonia oxidizing bacteria(AOB).Three treatments were compared,including conventional fertilization(NS),conventional fertilization plus rape straw(RS)and conventional fertilization plus broad bean straw(BS).The results showed that the effect of different types of straw returning on the Alpha diversity index of AOB community was stronger than that of AOA.For AOB community,compared with NS,straw returning(RS and BS)improved its diversity index,in which the diversity index of RS was higher than that of BS.Veen analysis showed that compared with AOA,the number and distribution of OTU in AOB amoA gene sequence were significantly affected by different types of straw returning, in which the effect of RS was stronger than that of BS on OTUdistribution. Different types of straw returning had no significant effect on the species composition and relative abundance ofAOA community on class level (p>0.05). While in AOB community, the relative abundance of unclassified_p__Proteobacteria of RS was 2 times higher than that of NS (p<0.05) and 1.45 times higher than that of BS (p>0.05). Comparedwith NS, RS reduced the relative abundance of norank_p__ammonia-oxidizing_bactera_ensemble, BS missed this species. Inconclusion, the response of AOB to different types of straw returning was stronger than that of AOA. Compared with BS, RShad a stronger effect on the microbial community structure of AOB.
