摘要
为探索LGALS1对猪繁殖与呼吸综合征病毒(PRRSV)在Marc-145中复制的影响,设置LGALS1免疫血清和PRRSV共同孵育Marc-145细胞的试验组、LGALS1 siRNA转染Marc-145细胞后再接种PRRSV的试验组以及PRRSV直接孵育Marc-145细胞的对照组,利用间接免疫荧光、TCID50测定、荧光定量和Western blot来测定病毒的增殖、滴度变化和表达情况。结果发现,抗LGALS1血清组、转染LGALS1 siRNA质粒组与对照组荧光、TCID50、病毒mRNA和蛋白水平差异均不显著。结果表明,LGALS1对PRRSV在Marc-145中复制无影响。
To explore the effect of LGALS1 on replication of porcine reproductive and respiratory syndrome virus(PRRSV) in Marc-145,the LGALS1 immune serum and PRRSV were used to co-incubate the Marc-145 cells,the LGALS1 siRNA was transfected into Marc-145 cells,and then the PRRSV-injected test group and the PRRSV-inoculated Marc-145 cells were directly incubated,virus proliferation,titer changes and expression were determined by indirect immunofluorescence,TCID50 assay,fluorescence quantification and Western blot.The results showed that there was no significant difference in fluorescence,TCID50,viral mRNA and protein levels among the anti-LGALS1 serogroup,the transfected LGALS1 siRNA plasmid group and the control group.It was initially concluded that LGALS1 had no effect on PRRSV replication in Marc-145.
