摘要
Ⅲ型干扰素(IFN)又被称为IFN-λ,具有与Ⅰ,Ⅱ型IFN类似的抗病毒特性,但利用不同的配体-受体系统,主要在一些上皮细胞及免疫细胞上发挥作用。本研究评价了用昆虫杆状病毒表达系统制备的重组鸡IFN-λ(chIFN-λ)在体内、外对禽Ⅰ群腺病毒4型(FAdV-4)GX2013株和8b型(FAdV-8b)QD2016株感染的抗病毒效果。在体外抗病毒试验中,重组chIFN-λ在LMH细胞上对GX2013株和QD2016株的抗病毒效价分别达到1.33×10^6,1.78×10^6 U/mL。在体内抗FAdV-4感染试验中,重组chIFN-λ治疗组在攻毒5 d后试验鸡存活率达60%,而未进行治疗的对照组3 d内死亡率为100%。在体内抗FAdV-8b感染试验中,重组chIFN-λ治疗组和非治疗组试验鸡均无明显临床症状,但治疗组在攻毒5 d后呼吸道和消化道排毒水平均低于未治疗组,且差异极显著(P<0.01)。上述结果表明重组chIFN-λ作为一种新型的生物制剂,有望在禽腺病毒感染防控中发挥重要的作用。
TypeⅢinterferons is also known as IFN-λ,with similar antiviral properties with typeⅠand typeⅡinterferons,but using different ligand-receptor system.IFN-λmainly play a role in some epithelial cells and immune cells.In this study,the biological activity of recombinant chicken IFN-λ(chIFN-λ)prepared from the baculovirus expression system were evaluated in vitro and in vivo.The recombinant chIFN-λshowed significant antiviral effect on the infection of avian adenovirus type 4(FAdV-4)GX2013 strain and 8 b(FAdV-8 b)QD2016 strain.The antiviral titers of recombinant chIFN-λto GX2013 and QD2016 reached 1.33×10^6,1.78×10^6 U/mL on LMH cells respectively.In the in vivo FAdV-4 infection-treatment experiment,the survival rate of the recombinant chIFN-λtreated chickens was 60%after 5 days of challenge,compared with 100%in the untreated control group.In the in vivo FAdV-4 infection-treatment experiment,although there were no obvious clinical symptoms in both treated and non-treated chickens,the virus shedding levels from the respiratory and digestive tract in the treatment group were significantly lower than those in the untreated group after 5 days of treatment(P<0.01).The results suggest that recombinant chIFN-λhas a potential to be deveoped as a novel biological agent and may play an important role in the control of avian adenovirus infection.
