Expression of RAC1 gene in prehierarchical follicles of chicken ovaries

为探讨RAC1基因在鸡前等级卵泡的表达特性及定位,本试验以海兰褐蛋鸡为供试材料,采用半定量RT-PCR方法对RAC1基因在鸡前等级卵泡组织中的mRNA表达水平进行测定分析,并进一步采用免疫组化的方法对RAC1蛋白在前等级卵泡的表达模式进行蛋白质定位研究。结果表明,RAC1基因在鸡不同直径的前等级卵泡内均有表达,但表达丰度存在一定的差异,在直径6~8 mm卵泡中相对表达量显著高于直径4~5.9,1~3.9 mm的卵泡(P<0.05);RAC1蛋白主要在卵母细胞、颗粒细胞和膜层细胞中表达,且主要分布在细胞质中,为进一步研究RAC1在鸡前等级卵泡发育中的作用提供了理论基础。

To investigate the expression of RAC1 gene and cellular localization in prehierarchical follicles of chicken ovaries,the expression abundances of RAC1 in prehierarchical follicles sampled from Hylan brown layers were determined by using by semi-quantitative RT-PCR,and the localization of RAC1 protein in prehierarchical follicles was further examined by immunohistochemistry in this study.The results showed that RAC1 gene was expressed in prehierarchical follicles with different diameters in hens ovaries,but the expression abundance was different to some extent.The relative expression of RAC1 gene in follicles with diameters of 6-8 mm was significantly higher than that in follicles with diameters of 4-5.9 mm and 1-3.9 mm(P<0.05).RAC1 protein was dominantly expressed in oocytes,granulosa cells and membrane cells,and was mainly distributed in cytoplasm.The results laid a foundation for further study on the role of RAC1 in the development of prehierarchical follicles in chickens.
