摘要
目的探讨当归芍药散对小胶质细胞活化状态的影响。方法采用CCK-8法和实时荧光定量PCR(Real-time PCR)检测不同浓度的当归芍药散对小胶质细胞活力的影响,及其对小胶质细胞不同活化状态生物标志物表达的影响。结果当归芍药散可以促进小胶质细胞的增殖,且呈现时间、剂量依赖性;当归芍药散可下调TNF-α mRNA的表达水平,同时上调Arg1 mRNA的表达水平。结论当归芍药散可促进小胶质细胞的增殖,并调控其活化状态向M2型转化,促进其对Aβ的吞噬作用从而改善阿尔兹海默病。
Objective To investigate the effect of Danggui Shaoyao San(DSS)on the microglia activation.Methods CCK-8 and Real-time PCR were used to detect the effects of DSS at different concentrations on the viability of microglia and their effects on the expression of biomarkers in different activation states of microglia.Results DSS can promote the proliferation of microglia in a time-and dose-dependent manner.DSS can down-regulate the expression of TNF-αmRNA and up-regulate the expression of Arg1 mRNA.Conclusion DSS can promote the proliferation of microglia and regulate its activation status to M2 type,and eventually promote its phagocytesis of Aβto improve Alzheimer’s Disease.
