辣木叶高效高压差低温连续式提取工艺优化及过程控制指纹图谱研究

Optimization of High Performance High Pressure and Low Temperature Successive Extraction for Moringa Oleifera Leaves and HPLC Fingerprint Analysis of Process Control

摘要 目的优选辣木叶提取工艺。方法采用高效液相色谱(HPLC)指纹图谱,对比热回流提取法和高效高压差低温连续式提取法(HHLSE)的提取效率;以蛋白质提取率为评价指标,通过正交试验设计进一步优化HHLSE提取工艺参数;以没食子酸和异槲皮苷为指标成分,利用HPLC指纹图谱追踪HHLSE工艺中提取、微滤、浓缩及喷雾过程化学成分的转移,分析高压差提取工艺优势。结果 HHLSE提取最佳工艺为:提取溶剂水,溶剂倍数为30倍,蛋白质提取率达48.56%。HPLC指纹图谱结果显示,HHLSE过程中,药材中没食子酸和异槲皮苷等有效成分均高效转移至最终提取物。结论优选的HHLSE提取工艺高效、稳定、简单,可为辣木叶相关产品的开发提供可靠的提取工艺。 Objective To optimize the extraction process of Moringa oleifera leaves. Methods The extraction rate were used to compare thermal recycling extraction and high-efficiency, high-pressure differential and low temperature continuous extraction(HHLSE) through HPLC fingerprint analysis. Taking the extraction rate of protein as evaluation index, orthogaonal test was used to further optimize process parameters of the HHLSE extraction technology. With gallic acid and isoquercetin as index ingredient of HPLC fingerprint, chemical composition of Moringa oleifera leaves was tracked in the extraction, microfiltration, concentration and spray process of the HHLSE, to analyze advantage of the HHLSE extraction technology. Results The optimal parameters of the HHLSE extraction process was as follows: 1:30(g/ml) ratio of dry power to water, with the protein extraction rate up to 48.56 %. The results of HPLC fingerprints showed that the effective components such as gallic acid and isoquercetin in the raw materials were transferred to the final extract efficiently during the HHLSE. Conclusion The HHLSE extraction process is efficient, stable and simple, which can provide a reliable extraction process for product development of Moringa oleifera leaves.
