黄连单枝连商品规格等级及生物碱含量研究

Study on commodity specification standard and alkaloids content of Coptis chinensis

目的研究黄连药材单枝连的外观性状量化指标以及与药材内在生物碱含量的相关性,为黄连单枝连药材商品规格等级的合理划分提供依据。方法对不同产地单枝连外观性状特征进行聚类分析。采用HPLC法测定了单枝连中6种生物碱含量,并对外观性状指标与内在生物碱含量之间进行了相关性分析,结合生产实际,依据分析结果建立单枝连商品规格分级标准。结果结合生产实际筛选出单枝连长、直径和单枝重作为分级指标,通过聚类分析,将黄连单枝连商品规格分为四个等级。特级黄连长不小于4.5 cm,直径不小于7.0 mm,重量不低于1.4 g;Ⅰ级黄连长不小于4.0 cm,直径不小于5.5 mm,重量不低于1.1 g;Ⅱ级黄连长不小于3.5 cm,直径不小于4.5 mm,重量不低于0.8 g;Ⅲ级黄连长不小于2.5 cm,直径不小于3.5 mm,重量不低于0.5 g。生物碱含量测定结果表明不同商品等级黄连的生物碱含量间无显著差异,不同产地黄连生物碱含量存在显著差异。相关性分析表明,单枝连外观性状指标与内部生物碱含量无显著相关性。结论此方法制定出的黄连商品规格分级标准适用于黄连商品生产流通领域,有利于黄连药材的质量控制。

Objective To establish a quality classification standard for Coptis chinensis. Methods The 24 areas medicinal materials of Coptis chinensis were respectively measured in quantitative chracteritics of appearance and the alkaloids content. Then the determination data were evaluated by spss correlation analysis and cluster analysis. Results Combining with the actual production, the commodity grade standard of Coptis chinensis was formulated.Correlation analysis indicated there was no significant correlation between quantitative chracteritics of appearance and the alkaloids content. Using cluster analysis, combining actual production,length, diameter and weight of single root were filtrated as classification index and the commercial specification grading of Coptis chinensis was divided into 4 grades. The primary quality classification standard of commercial specification for Coptis chinensis was established.The medicinal material quality of each grade should reach following requirements: for special grade medicinal material,length≥4.5 cm, diameter≥7.0 mm, weight≥1.4 g;for first grade seeds, length≥4.0 cm, diameter≥5.5 mm, weight≥1.1 g;for second grade seeds, length≥3.5 cm, diameter≥4.5 mm, weight≥0.8 g;for third grade seeds, length≥2.5 cm, diameter≥3.5 mm, weight≥0.5 g. Conclusion This classification standard of commercial specification of Coptis chinensis can be used as the quality control standard of Coptis chinensis in market.
