芪蛭降糖片对糖尿病大鼠胰岛损伤的保护机制

摘要 〔目的〕观察芪蛭降糖片对链脲佐菌素诱导糖尿病大鼠胰岛损伤的保护作用,探索其可能的作用机制。〔方法〕用链脲佐菌素诱导大鼠糖尿病模型,模型建立后采用人体等效日剂量的芪蛭降糖片、盐酸二甲双胍片和格列本脲片每日1次独立治疗3 mon,观察大鼠糖尿病胰岛损伤的动态变化,检测血糖、血清胰岛素(IMS)、糖化血清蛋白(GSP)及诱导型一氧化氮合酶(iNOS),网络药理学分析芪蛭降糖片保护糖尿病胰岛损伤的机制。〔结果〕与模型大鼠相比,盐酸二甲双胍片和格列本脲片虽对高血糖、多饮多食多尿、消瘦和血清低胰岛素有改善作用,对血清iNOS、GSP和胰腺组织损伤改善作用不明显。芪蛭降糖片降糖作用不及盐酸二甲双胍片和格列本脲片起效快,但改善多饮多食多尿、消瘦和血清低胰岛素方面与二者相似,且明显促进胰腺组织损伤修复。网络药理学分析表明,芪蛭降糖片通过PI3K-Akt信号通路、胰岛素信号通路、胰岛素抵抗等调节胰岛素分泌、炎症反应等生物过程来阻止胰腺损伤。〔结论〕芪蛭降糖片可以促进糖尿病大鼠胰腺损伤修复,是应用前景较好的糖尿病胰岛损伤防治药物。 〔Objective〕To observe the protective action of Qizhi hypoglycemic tablets on pancreatic islet injury in streptozotocin-induced diabetic rats and explore its possible mechanism.〔Methods〕Streptozotocin was used to prepare diabetic model in rats.After the diabetic model succeed,human equivalent daily dose of qizhi hypoglycemic tablets,metformin hydrochloride tablets and glibenclamide tablets alone via intragastric administration were treated independently once a day for three months,and the dynamic changes in islet injury were investigated in diabetic rats,the examined index included blood glucose,serum levels of insulin(INS),glycated serum protein(GSP)and inducible nitric oxide synthase(iNOS).Network pharmacology was used to analyze protective mechanism.〔Results〕Compared with model rats,Metformin hydrochloride tablets and glibenclamide tablets significantly improved hyperglycemia,polydipsia,polyphagia,polyuria,emaciation and the low serum insulin.There was no significant improvement on the level of iNOS,GSP and pancreatic tissue injury.Qizhi hypoglycemic tablets showed a similar effect on polydipsia,polyphagia,polyuria,emaciation and the low serum insulin to metformin hydrochloride tablets and glibenclamide tablets,although it was inferior to them in hypoglycemic action.Importantly,Qizhi hypoglycemic tablets significantly promoted pancreatic injury repair.Network pharmacology analysis shows that Qizhi hypoglycemic tablets can prevent pancreatic injury by regulating biological secretion such as“PI3K-Akt signaling pathway”,insulin signaling pathway,insulin resistance and other biological processes.〔Conclusion〕Qizhi hypoglycemic tablets can promote pancreatic injury repair and is a better preventive and therapeutic drug for diabetic islet injury.
