不同干燥方法对金盏银盘中苯丙素类成分含量的影响 被引量:1

摘要 目的建立高效液相色谱法同时测定鲜品金盏银盘中绿原酸、新绿原酸和隐绿原酸3种苯丙素类成分含量,并考察阴干、烘干、晒干及冻干等4种不同干燥方法对这3种成分含量的影响。方法采用反相高效液相色谱法,色谱柱为Welch Materials Ultimate XB C18(250 mm×4.6 mm,5μm),体积分数为0.1%的甲酸水溶液-乙腈为流动相,梯度洗脱,流速为1.0 mL·min^-1,检测波长为280 nm,柱温为30℃。鲜品金盏银盘采用不同方法干燥后,水加热回流提取,经处理后测定。结果所建立的HPLC方法可用于金盏银盘中3种苯丙素类成分的含量测定;在不同方法制备的样品中,鲜品含量最高,其次为冷冻干燥、晒干、阴干,烘干样品含量最低。结论该方法可用于考察不同干燥方法对金盏银盘主要成分含量的影响,并为其质量控制提供参考依据。 Objective To develop an HPLC method for determination of chlorogenic acid,neochlorogenic acid and cryptochlorogenic acid in Bidens biternata(Lour.)Merr.et Sherff herbs,to evaluate the effect of four drying methods(shade drying,oven drying,sun drying and vacuum freeze drying)on contents of three constituents.Methods The Welch Materials Ultimate XB C18(4.6 mm×250 mm,5μm)column was used with gradient elution of acetonitrile-0.1%formic acid solution at a wavelength of 280 nm.The flow rate was 1.0 mL·min-1 and column temperature was at 30℃.The fresh herb which was dried by different ways was determined after hot water reflux extraction.Results The established method could be used for content determination of three constituents in B.biternata(Lour.)Merr.et Sherff.The fresh herb among the samples prepared by different methods had the highest content,then followed by vacuum freeze drying,sun drying and shade drying,while the lowest was in oven drying.Conclusion The established method with a high sensitivity and good reproducibility can be used for evaluate the effect of the determination of major constituents in herbs by different drying methods,and provide a reference for its quality control.
