GSDMD及异构体在皮肤黑色素瘤的表达及预后研究

Study on the expression and prognosis of GSDMD and its isomers in cutaneous melanoma

目的探讨消皮素D(GSDMD)基因及其异构体在皮肤黑色素瘤中的表达及预后意义。方法应用基因表达谱数据动态分析(GEPIA)在线工具分析来自癌症基因组图谱(TCGA)数据库和基因型-组织表达(GTEx)项目中各类肿瘤及对应正常组织GSDMD表达情况,筛选差异表达有统计学意义的癌种。同时分析GSDMD与各类肿瘤的预后的相关性,筛选有预后相关性的癌种。进一步分析GSDMD表达与皮肤黑色素瘤分期的关系,及其异构体与皮肤黑色素瘤预后的关系。然后从TCGA数据库下载皮肤黑色素瘤(RNA-seq)基因表达数据,对GSDMD基因高低表达组间的差异基因进行基因本体(GO)富集及京都基因与基因组百科全书(KEGG)富集分析。结果GSDMD基因在皮肤黑色素瘤中高表达(P<0.05),且GSDMD与皮肤黑色素瘤患者的OS及DFS具有相关性(P<0.05),其中异构体002、004、005、007、011、012的表达与预后相关(P<0.05)。富集分析表明差异基因主要集中于免疫和炎性反应相关的功能集及信号通路。结论GSDMD基因在皮肤黑色素瘤中高表达,且与患者预后相关。

Objective The aim of the present study was to investigate the expression of Gsdermin D(GSDMD)in cutaneous melanoma and to evaluate its prognostic significance.Methods Gene Expression Profiling Interactive Analysis(GEPIA)online tool was used to analyze the expression of GSDMD in all kinds of tumors and corresponding normal tissues from The Cancer Genome Atlas(TCGA)database and The Genotype-Tissue Expression(GTEx)project.At the same time,the correlation between GSDMD and the prognosis of all kinds of tumors was analyzed,and the cancer species with prognosis correlation were screened.The relationship between the expression of GSDMD and the stage of cutaneous melanoma,and the relationship between its isomer and the prognosis of cutaneous melanoma were analyzed.Then we download the gene expression data of skin melanoma(RNA-seq)from TCGA database,and analyze the differential expression genes related to GSDMD gene expression.Furthermore,Gene Ontology(Go)enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment were analyzed.Results GSDMD gene was highly expressed in cutaneous melanoma(P<0.05).GSDMD was correlated with OS and DFS in patients with cutaneous melanoma(P<0.05),and the expression of isomers 002,004,005,007,011,012 was correlated with prognosis(P<0.05).Enrichment analysis showed that the differentially expressed genes mainly focused on the functional set and signal pathway related to immune and inflammatory response.Conclusion GSDMD gene is highly expressed in cutaneous melanoma,and it is related to the prognosis of patients.
