摘要
为工业化控制黄曲霉毒素B 1(aflatoxin B 1,AFB 1)污染,筛选吸附率高、稳定性好、脱毒后具备食品安全的优势菌株,应用超高效液相色谱-三重四级杆串联质谱(ultra performance liquid chromatography-tandem mass spectrometry,UPLC-MS/MS)技术检测菌株发酵液AFB 1残留量的方法,从35份采集自甘肃不同地域的西北酸菜样品中筛选到1株活菌脱毒率为66.41%、解吸后仍保留脱毒率为53.7%的菌株L34-2。菌株L34-2经形态学、生理生化及16S rRNA基因序列扩增测序,鉴定为布氏乳杆菌(Lactobacillus buchneri)。通过磷酸缓冲盐溶液(phosphate buffer saline,PBS)洗脱,体外模拟胃液消化,研究菌株-AFB 1复合体稳定性,结果表明,菌株L34-2吸附AFB 1具有较好的稳定性。
In order to solve the problem of industrial large-scale application of biological method to remove aflatoxin B 1(AFB 1),an AFB 1-adsorptive strain L34-2 was isolated from pickled vegetable in northwest China and its AFB 1 detoxification rate was 66.41%.Based on the morphological,physiological,biochemical analysis and sequence alignment of 16S rRNA,it was identified as Lactobacillus buchneri.The stability of the bacteria-AFB 1 complex was studied by phosphate buffer saline(PBS)elution and in vitro gastric juice digestion simulation.The results showed that it had good stability in adsorption of AFB 1.
