响应曲面法优化壁虎水溶性蛋白的提取工艺 被引量:1

摘要 目的:优化壁虎中水溶性蛋白的提取工艺。方法:在单因素提取试验的基础上,以pH、提取温度、提取时间、料液比为自变量,蛋白质提取含量为指标,通过Box-Behnken响应曲面法优化中药壁虎中水溶性蛋白的超声提取工艺。结果:优化得到壁虎水溶性蛋白的最佳提取工艺为料液比1∶70,提取时间84 min,pH 14,此工艺条件下壁虎水溶性蛋白质质量分数为272.92 mg·g^-1。结论:该优化工艺准确可靠,且具有一定的可操作性和实用价值。 Objective:To explore the optimum extraction process of water soluble protein from gekko.Methods:Based on single factor test,pH value,extraction time,extraction temperature,solid-liquid ratio were selected as independent variables,the extracting of protein content was used as dependent variables,Response surface methodology of Box-Behnken Design was applied to optimize the ultrasonic extraction conditions of water soluble protein from gekko.Results:The optimal extraction conditions were determined as follows:liquid-soild ratio of 70∶1,pH 14,ultrasonic extraction time 84 min,the extraction content of water soluble protein could be reached 272.92 mg·g^-1 under the optimal conditions.Conclusion:The optimized process is accurate,reliable,operable and practical.
