Extraction Optimization and Determination of Content of Total Flavonoids in 13 Batches of Different Parts of Cissus assamica

目的:优化红背丝绸总黄酮的提取工艺,并建立其药材的提取和含量测定方法。方法:采用回流提取法提取红背丝绸总黄酮,通过单因素试验,考察提取方法、提取溶剂浓度、提取体积、提取时间对药材总黄酮含量的影响,采用正交试验,优化提取工艺条件,筛选出红背丝绸总黄酮的最佳提取工艺。结果:红背丝绸总黄酮的最佳提取工艺为70%乙醇,料液比为1∶30,回流提取时间为1.0 h。结论:该方法能有效测定红背丝绸总黄酮的含量,重复性较好、方法稳定可行,为该药材的质量标准研究提供参考。

Objective:To optimize the extraction process of total flavonoids from leaves of Cissus assamica and establish a method for the extraction and determination of total flavonoids.Methods:The reflux extraction method was used to extract total flavonoids from the leaves of C.assamica.The effects of extraction method,solvent concentration,extraction volume and extraction time on the content of total flavonoids in C.assamica were investigated by single factor test.The orthogonal test was used to optimize the extraction conditions and the optimum extraction process was selected.Results:The optimum extraction process of total flavonoids from the leaves of C.assamica was 70%ethanol,the ratio of material to liquid was 1∶30,and the reflux extraction time was 1.0 h.Conclusion:The method can effectively determine the content of total flavonoids in the leaves of C.assamica,and provide a scientific basis for the quality standard research of the herb.The extraction of total flavonoids from the leaves of C.assamica has good repeatability and stability.
