摘要
目的:探讨半夏泻心结肠靶向片对溃疡性结肠炎(UC)模型大鼠的治疗作用。方法:采用二硝基苯磺酸(DNBS)-乙醇法建立UC大鼠模型,除正常组外,其余大鼠均造模,模型建立后将大鼠随机分为模型组、美沙拉嗪组和半夏泻心高、中、低剂量组。连续给药14 d后,观察各组大鼠结肠组织形态并测量血清细胞因子的变化。结果:除模型组大鼠大便松散有隐血外,其余各组恢复较好。美沙拉嗪组和半夏泻心高剂量组大鼠第15天DAI评分、结肠CMDI评分和血清TNF-α、IL-8水平明显低于模型组(P<0.05或P<0.01),而IL-4水平明显高于模型组(P<0.01);半夏泻心高剂量组大鼠以上指标与美沙拉嗪组比较,差异均无统计学意义(P>0.05)。光镜下各给药组大鼠结肠组织仍有不同程度的炎性浸润,半夏泻心高剂量组大鼠症状最轻。结论:高剂量的半夏泻心结肠靶向片能显著改善UC大鼠结肠组织修复及调节炎性因子水平,治疗效果趋近美沙拉嗪。
Objective:To investigate the therapeutic effect of Banxia Xiexin colon targeted tablets on ulcerative colitis(UC)model rats.Methods:The UC rat model was established by dinitrobenzene sulfonic acid(DNBS)-ethanol method.Except for the normal group,all other rats were modeled.After the model was established,the rats were randomly divided into model group,mesalazine group and Banxia Xiexin high,medium and low dose groups.After 14 days of continuous administration,the changes of colonic tissue morphology and serum cytokines were observed.Results:Except for loose stool and occult blood in the model group,the other groups recovered well.The DAI score of day 15,colon CMDI score and serum TNF-αand IL-8 levels of the mesalazine group and Banxia Xiexin high-dose group were significantly lower than those in the model group(P<0.05 or P<0.01),while the IL-4 level was significantly higher than that in the model group(P<0.01).Meanwhile,there was no significant difference in the above indexes between the Banxia Xiexin high-dose group and the mesalazine group(P>0.05).Under the light microscope,the colon tissue of rats in each administration group still had different degrees of inflammatory infiltration,and the symptoms of the rats in the Banxia Xiexin high-dose group were the least.Conclusion:High-dose Banxia Xiexin colon targeted tablets can significantly improve the colon tissue repair and regulate the level of inflammatory factors in UC rats,and the therapeutic effect is similar to mesalazine.
