滇白珠醇提物预防大鼠胃溃疡的实验研究

摘要 目的:探讨滇白珠醇提物(DBZ)预防给药对大鼠幽门结扎型胃溃疡(A)及乙醇型胃溃疡(B)的抑制作用。方法:采用A模型,观察DBZ对大鼠胃溃疡发生率、溃疡指数以及胃液量、胃酸、胃蛋白酶的影响;采用B模型,观察DBZ对大鼠胃溃疡发生率、溃疡指数以及胃黏膜组织SOD、MDA、PGE2、NO含量的影响。结果:A模型为瘤胃部胃溃疡。模型组胃溃疡发生率高,溃疡指数大,病理检查显示溃疡部位胃组织严重缺损。与模型组比较,DBZ高剂量组大鼠胃溃疡发生率及溃疡指数显著减少,胃组织缺损程度明显减轻,胃液量、胃酸浓度和总酸排出量均显著降低;DBZ低剂量组大鼠胃溃疡指数显著减小,胃组织缺损程度明显减轻,胃酸浓度显著降低;DBZ高、低剂量组对胃蛋白酶活力均没有显著影响。B模型为腺胃部胃溃疡。模型组大鼠胃部出现明显淤血及溃疡。与模型组比较,DBZ高剂量组大鼠胃部没有淤血及溃疡,DBZ低剂量组淤血及溃疡程度明显减轻;DBZ高、低剂量组大鼠胃黏膜组织的SOD含量显著增加、MDA含量显著降低,而对PGE2、NO含量没有显著影响。结论:DBZ预防给药对大鼠A、B模型胃溃疡均具有显著抑制作用。其中对A模型抑制作用可能与减少胃酸分泌有关,对B模型抑制作用可能与升高胃黏膜SOD水平、同时降低MDA水平有关。 Objective: To investigate the inhibitory effect of the preventive administration of Dianbaizhu ethanol extract(DBZ) on pyloric ligated gastric ulcer(A) and ethanolic gastric ulcer(B) in rats. Methods: Model A was used to observe the effect of DBZ on the incidence of gastric ulcer, ulcer index, gastric fluid volume, gastric acid and pepsin. Model B was used to observe the effect of DBZ on the incidence of gastric ulcer, ulcer index and the content of SOD, MDA, PGE2 and NO in gastric mucosa. Results: Model A was rumen gastric ulcer. In the model group, the incidence of gastric ulcer was high and the ulcer index was large. Pathological examination showed that the gastric tissue at the ulcer site was seriously damaged. Compared with the model group, the incidence of gastric ulcer and ulcer index in the DBZ high-dose group were significantly reduced,the degree of gastric tissue defect was significantly reduced, and the amount of gastric fluid, gastric acid concentration and total acid output were significantly reduced. In the DBZ low-dose group, the gastric ulcer index of rats was significantly reduced, the degree of gastric tissue defect was significantly reduced, and the concentration of gastric acid was significantly reduced. The DBZ high and low dose groups had no significant effect on pepsin activity. Model B was gastric ulcer in the glandular stomach. Compared with the model group, there was no congestion and ulcer in the stomach of the DBZ high-dose group, and the degree of congestion and ulcer in the DBZ low-dose group was significantly reduced. Meanwhile, in the DBZ high and low dose groups, the SOD content in the gastric mucosa tissue of the rats was significantly increased, and the MDA content was significantly reduced, but there was no significant effect on the content of PGE2 and NO. Conclusion: Prophylactic administration of DBZ has a significant inhibitory effect on gastric ulcers in rats A and B models. The inhibitory effect on model A may be related to the reduction of gastric acid secretion, and the inhibitory effect on model B may be related to the increase of gastric mucosal SOD level and the decrease of MDA level.
