基于网络药理学探究黄连生物碱干预原发性高血压的作用机制 被引量:3

摘要 目的:基于网络药理学探讨黄连生物碱对原发性高血压的作用机制。方法:采用TCMSP、PubChem、BATMAN数据库等,以口服生物利用度和类药性为限制条件筛选黄连化学成分;通过PubChem数据库筛选符合异喹啉型的生物碱化合物,通过Uniprot数据库获取黄连生物碱的作用靶点,构建黄连生物碱作用靶点的蛋白相互作用网络;基于GeneCards数据库筛选原发性高血压的相关靶点,利用Venny2.1工具绘制黄连生物碱作用靶点和高血压疾病靶点的韦恩图,通过生物信息学手段对黄连生物碱对高血压的作用靶点进行GO分析及KEGG分析。结果:富集通路分析KEGG通路有神经活性配体受体相互作用、cAMP信号通路、钙信号通路、间隙连接、HIF-1信号通路、cGMP-PKG信号通路、多巴胺能突触信号通路等。结论:黄连生物碱通过调节神经机制、离子浓度、抗氧化应激、抗炎等相关机制干预高血压。 Objective:To explore the effect mechanism of Coptis Alkaloid on essential hypertension based on network pharmacology.Methods:TCMSP,PubChem,BATMAN databases,etc.were used to screen the Chemical constituents of Huanglian(Coptis chinensis)with oral bioavailability and drug-like properties as the limiting conditions;The isoquinoline alkaloids were screened by PubChem database,and the targets of Coptis Alkaloids were obtained according to UniProt database;The protein interaction network was established for the targets of Coptis Alkaloid.Primary target related targets for essential hypertension was screened by GeneCards and the a Wayne diagram of targets of Coptis Alkaloid and Essential hypertension was drawn with Venny2.1.GO analysis and KEGG analysis of the effects of Coptis Alkaloid on hypertension were performed by bioinformatics.Results:Enriched pathway analysis showed that KEGG pathway had neuroactive ligand receptor interactions,cAMP signaling pathway,calcium signaling pathway,gap junction,HIF-1 signaling pathway,cGMP-PKG signaling pathway,and dopaminergic synaptic signaling pathway.Conclusion:Coptis Alkaloid intervene in essential hypertension by regulating the neural mechanism,ion concentration,anti-oxidative stress and anti-inflammatory factors.
