染色体加倍对红掌组培快繁特性的影响 被引量:1

Effects of Chromosome Doubling on Micropropagation Charateristics of Anthurium ‘Pink Champion’

摘要 以二倍体、三倍体和四倍体‘粉冠军红掌’幼叶为外植体,研究了染色体加倍对红掌组培快繁特性的影响。结果表明:不同倍性红掌的幼叶起始脱分化时间相近,愈伤组织诱导率差异不显著,说明染色体加倍对红掌幼叶脱分化影响不显著;染色体加倍对红掌愈伤组织增殖影响显著,在MS+6-BA 1.0 mg·L^-1+NAA 0.2 mg·L^-1培养基上,二倍体增殖率最高,四倍体次之,三倍体最低;染色体加倍对红掌愈伤组织芽分化也有显著影响,三倍体的芽分化率显著低于二倍体和四倍体,二倍体、三倍体和四倍体愈伤组织芽分化的适宜外源激素浓度分别为6-BA 0.5 mg·L^-1+KT 0.2 mg·L^-1、6-BA 0.5 mg·L^-1+KT 0.5 mg·L^-1和6-BA 1.0 mg·L^-1+KT 0.2 mg·L^-1;染色体加倍对红掌根分化率和试管苗移栽成活率没有显著影响。上述研究结果表明,染色体加倍对红掌组培快繁效率影响显著,且不同倍性影响不同。 The effect of chromosome doubling on micropropagation characteristics of Anthurium’Pink Champion’was investigated by using of young leaves as explants.The results indicated that the initial dedifferentiation time of diploid,triploid and tetraploid Anthurium’Pink Champion’was similar,and the differences in leaf callus induction rate were not significant,suggested that chromosome doubling had little effect on leaf callus induction.Chromosome doubling significantly affected callus proliferation,on MS+6-BA 1.0 mg·L^-1+NAA 0.2 mg·L^-1 medium,the callus proliferation rate of diploid was the highest,followed by the tetraploid,the proliferation rate of triploid was the lowest.Chromosome doubling also had significant effect on bud differentiation,the bud differentiation rate of triploid was significantly lower than that of diploid and tetraploid,the suitable exogenous hormone concentrations for bud differentiation of diploid,triploid,tetraploid were 6-BA 0.5 mg·L^-1+KT0.2 mg·L^-1,6-BA 0.5 mg·L^-1+KT 0.5 mg·L^-1 and 6-BA 1.0 mg·L^-1+KT 0.2 mg·L^-1,respectively.Chromosome doubling had little influence on root differentiation rate and transplanting survival rate.The results obtained demonstrated that chromosome doubling had significant effect on micropropagation efficiency in Anthurium andraeanum,which depended on ploidy level.
