红豆杉活性成分筛选的一种新方法的研究 被引量:3

New screening method for active constituents in Taxus chinensis

摘要 目的:建立一种新型的药物活性成分筛选方法,用于筛选红豆杉抗肿瘤活性成分群。方法:在细胞正常代谢的基础上,对不同时间段、不同细胞模型的细胞上清液进行液相微萃取处理,对其中的活性成分群进行有效检测,根据色谱峰变化发现潜在活性成分群。将MCF-7、A549细胞作为活性筛选的平台,在细胞自然吸收状态下,以红豆杉提取物为研究对象,对中药材中的活性成分群进行筛选和捕获,而后进行HPLC分析。结果:成功筛选出红豆杉中的抗肿瘤活性成分群分别是脱乙酰巴卡亭、巴卡亭Ⅲ、紫杉碱、去乙酰紫杉醇、三尖杉宁碱、紫杉醇、金松双黄酮、云南紫杉烷。结论:本研究试图为活性成分的筛选、中药质量的综合评价提供一种方便、普适和高效的新方法,为中药药效物质基础研究提供理论基础和实验依据。 Objective:To establish a new method for screening antitumor active components in Taxus chinensis.Methods:On the basis of normal cell metabolism,the supernatant of cells in different time periods and cell models was treated with liquid phase microextraction,the active component groups were effectively detected,and the potential active component groups were found according to the changes of chromatographic peaks.MCF-7 and A549 cells were used as the platform for activity screening.Under the condition of natural absorption of cells,Taxus chinensis extract was used as the research object to screen and capture the active component groups in Chinese medicinal materials,and then HPLC analysis was performed.Results:10-deacetylbaccatinⅢ,baccatinⅢ,taxinine M,10-deacetyltaxol,cephalomannine,taxol,sciadopitysin and taxuyunnanine C were screened as the antitumor active components from Taxus chinensis.Conclusion:This study attempts to provide a convenient,universal and efficient new method for the screening of active ingredients and the comprehensive evaluation of the quality of traditional Chinese medicine,which provides the theoretical basis and experimental basis for the basic research on the medicinal substances of traditional Chinese medicine.
