黄蜀葵花总黄酮的质量标准及指纹图谱研究 被引量:4

摘要 目的对黄蜀葵花总黄酮进行定性定量实验研究,建立黄蜀葵花总黄酮的质量标准及HPLC指纹图谱,为该有效部位的制剂开发提供参考。方法采用化学鉴别法和薄层色谱法对黄蜀葵花总黄酮进行定性鉴别;采用醋酸-醋酸钠-氢氧化铝法进行黄蜀葵花总黄酮含量测定;采用HITACHI LaChrom C18色谱柱(250mm×4.6mm,5μm),以乙腈-0.1%磷酸溶液作为流动相进行梯度洗脱,柱温30℃,检测波长360nm,流速1.0mL·min-1,进样量10μL,对10个不同批次提取物进行测定;采用HPLC-DAD结合相似度评价建立黄蜀葵花总黄酮指纹图谱。结果拟定了黄蜀葵花总黄酮的质量标准草案,建立了黄蜀葵花总黄酮的指纹图谱,确定了11个共有峰。结论本研究建立的定性定量检测方法操作简便、准确可靠、专属性强、重复性好,可作为黄蜀葵花总黄酮的质量控制方法;建立的黄蜀葵花总黄酮HPLC指纹图谱可为该提取物的鉴别及质量控制提供参考。 OBJECTIVE The qualitative and quantitative experiments on total flavonoids of Abelmoschl manihot L medic flowers were carried out to establish the quality standard and HPLC fingerprint of the total flavonoids of Abelmoschl manihot L medic flowers,which provided a reference for the development of the effective part.METHODS First,the total flavonoids of Abelmoschl manihot L medic flowers were qualitative identified by chemical identification and thin-layer chromatography and determined by Acetic acid-Sodium acetate-Aluminum hydroxide method.The,10 different batches of extract were analyzed on a HITACHI LaChrom C 18 chromatographic column(250mm×4.6mm,5μm),gradient elution with mobile phase of acetonitrile-0.1%phosphoric acid solution and water flowing at 1.0mL·min-1.And the injection volume was set at 10μL per injection detecting at 360nm in 30℃.Last,HPLC-DAD combined with similarity evaluation was used to establish the fingerprint of the total flavonoids of Abelmoschl manihot L medic flowers.RESULTS A draft quality standard for the total flavonoids of Abelmoschl manihot L medic flowers was prepared,and the fingerprint of the total flavonoids of Abelmoschl manihot L medic flowers was established.It was confirmed that there were 11 common peaks in these 10 batches.CONCLUSION The established method in this study is simple,accurate,reliable,specific,and reproducible,which can be used as a quality control method for the total flavonoids of Abelmoschl manihot L medic flowers.The established HPLC fingerprint of the total flavonoids of Abelmoschl manihot L medic flowers can provide reference for the identification and quality control of the extract.
