新疆过敏性哮喘患儿FCER2、CRHR1及GLCCI1基因多态性分析 被引量:2

Research of FCER2,CRHR1 and GLCCI1 gene polymorphisms on children with allergic asthma of Xinjiang

摘要 目的分析低亲和力免疫球蛋白E受体2(FCER2)rs28364072位点、促肾上腺皮质激素释放激素受体1(CRHR1)rs37973位点及糖皮质激素诱导转录基因1(GLCCI1)rs242941位点基因型和等位基因在新疆维吾尔族、汉族及哈萨克族过敏性哮喘患儿中的差异性分布特征。方法用荧光染色原位杂交方法检测273例新疆过敏性哮喘患儿(其中维吾尔族96例,汉族118例,哈萨克族59例)的FCER2 rs28364072、GLCCI1 rs37973及CRHR1 rs242941基因多态性,并分析基因型频率和等位基因频率在不同民族患儿间是否存在差异性。结果新疆3个民族273例过敏性哮喘患儿间FCER2 rs28364072和GLCCI1 rs37973基因型频率及等位基因频率差异均无统计学意义(均P>0.05)。汉族患儿的CRHR1 rs242941 GG型基因型频率均显著高于哈萨克族和维吾尔族患儿,差异均有统计学意义(95.8%vs 84.7%,88.5%,均P<0.05)。结论在与吸入性糖皮质激素疗效的相关性研究中,需考虑CRHR1与民族因素的差异,可不用考虑FCER2、GLCCI1与民族差异性的因素。 Objective To analyze the difference distributions of low-affinity immunoglobulin E receptor 2(FCER2)rs28364072,glucocorticoid-induced transcription gene 1(GLCCI1)rs37973 and corticotropin-releasing hormone receptor 1(CRHR1)rs242941 in Uygur,Kazak and Han children with suspected allergic asthma of Xinjiang.Methods The polymorphisms of FCER2 rs28364072,GLCCI1 rs37973 and CRHR1 rs242941 genes in 273 children with allergic asthma in Xinjiang(including 96 Uygur,118 Han and 59 Kazak)were detected by fluorescence staining in situ hybridization,and the differences of genotype frequency and allele frequency among different nationalities were analyzed.Results There were no significant differences in FCER2 rs28364072 and GLCCI1 rs37973 genotype frequencies and allele frequencies among 273 children with allergic asthma in three nationalities in Xinjian(all P>0.05).The CRHR1 rs242941 GG genotype frequency of Han children was significantly higher than that of Kazakh and Uyghur children,and the differences were statistically significant(95.8%vs 84.7%,88.5%,all P<0.05).Conclusion In the study of the correlation with the efficacy of inhaled glucocorticoids,the difference between CRHR1 and ethnic factors needs to be considered instead of FCER2 and GLCCI1.
