槲皮素固体脂质纳米粒对Aβ25-35损伤的PC12细胞作用的研究

摘要 目的探讨槲皮素固体脂质纳米粒(Que-SLN)对Aβ25-35损伤的PC12细胞的作用。方法通过熔融超声法制备Que-SLN,以透析法考察体外释放特性。以噻唑蓝(MTT)法检测不同处理组及不同时间的细胞活力大小,以荧光探针标记法考察细胞摄取率。结果体外释放实验表明,Que-SLN在72 h内具有缓释作用,累计释放率达到(87.60±0.66)%。MTT结果显示,槲皮素及Que-SLN均能促进PC12的增殖,Que-SLN的促增殖作用优于槲皮素且作用范围更广。50,100,200μmol·L^-1 Que-SLN组的细胞的存活率分别为(116.79±2.75)%,(119.46±1.89)%,(107.55±3.17)%,与Aβ25-35组(59.83±0.70)%相比,差异有统计学意义(P<0.05)。50μmol·L^-1 Que在24,48 h对Aβ25-35损伤的PC12细胞的保护作用基本相同(P>0.05),而Que-SLN在48 h对细胞的保护作用为(80.14±3.96)%,明显优于24 h的(67.28±3.10)%,差异有统计学意义(P<0.01)。结论Que-SLN具有缓释作用,Que-SLN对正常的PC12细胞具有促增殖作用,对Aβ25-35诱导的PC12细胞损伤具有保护作用且更容易被细胞摄取。 Objective To discuss the influence of quercetin solid lipid nanoparticles(Que-SLN)on PC12 cells damaged by Aβ25-35.Methods Que-SLN was prepared by melt ultrasound,in vitro release characteristics of dialysate.MTT assay was used to detect the cell viability in different groups and difference times,the cell uptake rate was studied by fluorescence probe labeling.Results In vitro release test showed that Que-SLN had a sustained release effect within 72 h and the cumulative release rate was(87.60±0.66)%.MTT results showed that Que and Que-SLN could promote the proliferation of PC12,and the effect of Que-SLN was better than Que,with a wider range of action.The survival rates of cells in 50,100,200μmol·L^-1Que-SLN groups were(116.79±2.75)%,(119.46±1.89)%,(107.55±3.17)%,with significant difference compared with Aβ25-35 group,which was(59.83±0.70)%(P<0.05).The protective effect on PC12 cells injured by Aβ25-35 in 50μmol·L^-1Que at 24,48 h was almost the same(P>0.05),while that in Que-SLN at 24,48 h were(67.28±3.10)%,(80.14±3.96)%,with significant difference(P<0.01).Conclusion Que-SLN has a sustained-release effect,Que-SLN can promote the proliferation of normal PC12 cells and protect PC12 cells from Aβ25-35-induced injury,and is more easily absorbed by cells.
