姜黄素对肝纤维化小鼠的影响 被引量:10

Effect of curcumin on liver fibrosis mice

摘要 目的研究姜黄素对肝纤维化小鼠的治疗效果及对肝组织核因子E2相关因子2/血红素加氧酶-1(Nrf2/HO-1)通路的影响。方法将36只小鼠随机分为对照组(n=12)、模型组(n=12)及实验组(n=12)。模型组与实验组小鼠用腹腔注射40%四氯化碳(CCl4)花生油的方法构建肝纤维化小鼠模型,每周干预2次,连续干预8周;每次干预前,实验组小鼠灌胃姜黄素200 mg·kg^-1,模型组与对照组小鼠灌胃等量生理盐水。以放射性免疫法检测透明质酸(HA)、层粘连蛋白(LN)、Ⅳ型胶原(CⅣ)、Ⅲ型前胶原肽(PⅢP)含量;以蛋白质印迹(Wb)法检测肝组织Nrf2/HO-1通路相关蛋白表达。结果对照组、模型组及实验组小鼠血清HA水平分别为(106.31±6.24),(332.52±38.63),(119.87±10.21)ng·mL^-1;血清LN水平分别为(51.37±3.92),(138.56±11.42),(74.16±1.93)ng·mL^-1;血清CⅣ水平分别为(10.03±1.12),(96.45±6.38),(34.59±4.11)μg·L^-1;血清PⅢP水平分别为(14.11±0.52),(39.26±1.37),(21.21±1.02)μg·L^-1,差异均有统计学意义(均P<0.05)。结论姜黄素可有效抑制肝纤维化小鼠疾病进展,有效缓解肝纤维化损伤,其作用机制可能与上调肝组织Nrf2/HO-1通路信号转导相关。 Objective To explore the treatment effect of curcumin for liver fibrosis mice and influence on Nuclear factor E2 related factor 2/Heme oxygenase 1 pathways in liver tissue.Methods Thirty-six mice were divided randomly into control group(n=12),model group(n=12)and test group(n=12).Mice in model group and test group were built liver fibrosis mice model by intraperitoneal injection of 40%carbon tetrachloride(CCl4)peanut oil,2 times per week,for 8 weeks.Mice in test group were gavage with curcumin 200 mg·kg^-1 before intervention per time,mice in model group and control group were gavage with the same amount of normal saline.Serum contents of hyaluronic acid(HA),laminin(LN),collagen typeⅣ(CⅣ),precollagenⅢpeptide(PⅢP)were detected by radiological immunization;the liver tissues pathology morphology and the degree of liver fibrosis were observed by hematoxylin and eosin(HE)staining and Masson staining;Western blot(Wb)method was used to detect the liver tissues Nrf2/HO-1 pathways related protein expression.Results The serum HA levels in control group,model group and test group were(106.31±6.24),(332.52±38.63),(119.87±10.21)ng·mL^-1;serum LN levels were(51.37±3.92),(138.56±11.42),(74.16±1.93)ng·mL^-1;serum CⅣlevels were(10.03±1.12),(96.45±6.38),(34.59±4.11)μg·L^-1;serum PⅢP levels were(14.11±0.52),(39.26±1.37),(21.21±1.02)μg·L^-1,all with significant difference(all P<0.05).Conclusion Curcumin can inhibit the disease progression of liver fibrosis mice,alleviate the injury of liver fibrosis effectively,the mechanism may be related to the up-regulation of liver tissue Nrf2/HO-1 signal transduction.
