葛根素联合雌二醇对大鼠骨质疏松性骨折愈合的影响 被引量:5

Effect of puerarin combined with estradiol on the healing of osteoporotic fracture in rats

摘要 目的探讨葛根素联合雌二醇对大鼠骨质疏松性骨折(OPF)愈合的影响。方法用双侧卵巢摘除术+骨折内固定术构建OPF模型,并随机分为模型组、对照A组、对照B组和实验组,每组20只;另取20只大鼠仅切除部分脂肪组织作为空白组。于造模成功后第2天,空白组和模型组均给予5 mL·kg-10.9%NaCl,皮下注射,qd;对照A组给予35 mg·kg^-1葛根素,皮下注射,qd;对照B组给予150μg·kg-1雌二醇,皮下注射,bid;实验组给予35 mg·kg^-1葛根素,皮下注射,qd+150μg·kg-1雌二醇,皮下注射,bid。5组大鼠均连续给药6周。用放射性X线片评估骨折愈合,用双能X线吸收扫描仪测量骨痂骨密度(BMD),用酶联免疫吸附实验法检测骨代谢指标。结果模型组、对照A组、对照B组和实验组的骨折愈合评分分别为(2.35±0.10),(3.80±0.14),(3.75±0.14)和(4.68±0.05)分,骨痂BMD分别为(0.10±0.01),(0.15±0.01),(0.16±0.01)和(0.19±0.01)g·cm^-2,血清钙分别为(1.92±0.10),(2.40±0.09),(2.38±0.12)和(2.50±0.10)mmol·L^-1,血清磷分别为(2.33±0.14),(3.30±0.13),(3.41±0.11)和(3.55±0.10)mmol·L^-1,血清碱性磷酸酶分别为(178.28±10.81),(152.75±9.34),(148.51±8.92)和(130.20±8.40)U·L^-1,实验组的上述指标与模型组比较,差异均有统计学意义(均P<0.05)。结论葛根素联合雌二醇可调节大鼠骨质疏松性骨折骨代谢,提高骨密度,加快骨折愈合。 Objective To explore the effect of puerarin combined with estradiol on the healing of osteoporotic fracture(OPF)in rats.Methods The bilateral oophorectomy+fracture internal fixation was used to build the OPF model.The model rats were randomly divided into model,control A,control B and experimental groups with 20 rats per group.Another 20 rats with part of the adipose tissue removed were selected as the blank group.Two days after successful modeling,blank and model groups were given 5 mL·kg-10.9%NaCl,hypodermic injection,qd;control A group was given 35 mg·kg^-1 puerarin,hypodermic injection,qd;control B group was given 150μg·kg^-1 estradiol,hypodermic injection,bid;experimental group was given 35 mg·kg^-1 puerarin,hypodermic injection,qd+150μg·kg^-1 estradiol,hypodermic injection,bid.Five groups were treated for 6 weeks.The fracture healing was measured by radiological X-ray.The epiphyseal bone mineral density(BMD)was measured by dual-energy X-ray absorption scanner.The bone metabolism was measured by enzyme-linked immunosorbent assay.Results In the model,control A,control B and experimental groups,the fracture healing scores were(2.35±0.10),(3.80±0.14),(3.75±0.14)and(4.68±0.05),the epiphyseal BMD were(0.10±0.01),(0.15±0.01),(0.16±0.01)and(0.19±0.01)g·cm^-2,the serum calcium were(1.92±0.10),(2.40±0.09),(2.38±0.12)and(2.50±0.10)mmol·L^-1,the serum phosphorus were(2.33±0.14),(3.30±0.13),(3.41±0.11)and(3.55±0.10)mmol·L^-1,the serum alkaline phosphatase were(178.28±10.81),(152.75±9.34),(148.51±8.92)and(130.20±8.40)U·L^-1.There were significant differences in the above-mentioned indexes between experimental group and model group(all P<0.05).Conclusion Puerarin combined with estradiol can regulate bone metabolism,increase bone mineral density and accelerate fracture healing in rats with osteoporotic fracture.
