摘要
目的:探索EP受体拮抗剂AH6809对炎症导致的BV-2小胶质细胞凋亡的影响。方法:将脂多糖加入体外培养BV-2细胞中制备炎症模型,通过倒置显微镜观察不同程度EP受体拮抗剂AH6809干预BV-2细胞的形态学变化,应用免疫荧光染色、流式细胞术、TUNEL染色与免疫印迹检测BV-2细胞后凋亡的情况。结果:AH6809浓度<80μmol/L时,BV-2细胞炎症反应减轻,无凋亡产生;AH6809浓度≥80μmol/L时,BV-2细胞出现凋亡;AH6809浓度达160μmol/L时,BV-2细胞凋亡数显著增多。结论:在脂多糖诱导BV-2细胞的炎症实验中,AH6809抗炎症反应的安全浓度控制在80μmol/L范围内。
Objective:To explore the effects of EP receptor antagonist AH6809 on the apoptosis of BV-2 microglial cells caused by inflammation.Methods:Lipopolysaccharide was added to the BV-2 cells in vitro to establish inflammation model and the morphological changes of BV-2 cells in different concentration of AH6809 intervention were observed by inverted microscope.Immunofluorescence staining,flow cytometry,TUNEL staining and Western blotting were used to detect the apoptosis of BV-2 cells.Results:When the concentration of AH6809 was<80μmol/L,the inflammatory response of BV-2 cells was alleviated but no apoptosis occurred.When the concentration of AH6809 was≥80μmol/L,BV-2 cells began to undergo apoptosis;when the concentration of AH6809 reached 160μmol/L,the number of apoptotic BV-2 cells increased significantly and reached a peak.Conclusion:In the lipopolysaccharide-induced inflammation experiment of BV-2 cells,the safe concentration of AH6809 for anti-inflammatory reaction is in the range of 80μmol/L.
