摘要
目的基于网络药理学方法联合GEO芯片差异基因分析,探析培土清心方治疗特应性皮炎的分子机制。方法检索GEO数据库,获取特应性皮炎的基因芯片,绘制差异基因火山图与热图。运用TCMSP、BATMAN-TCM、Drugbank数据库,获取培土清心方治疗特应性皮炎的有效成分及基因,并利用cytoscape 3.7.2,构建培土清心方-特应性皮炎-分子基因网络,行拓扑网络分析。运用R软件,对基因进行GO功能与KEGG通路富集分析。结果获得培土清心方治疗特应性皮炎的143个活性成分,治疗特应性皮炎的主要活性成分为槲皮素、豆甾醇、木犀草素。培土清心方对应疾病的核心基因共42个,其中重要候选基因为PPARG等。拓扑网络分析重要候选基因为TP53、NTRK1等。GO分析生物通路共516条,细胞成分8个,分子功能44个,主要涉及response to steroid hormone生物过程,nuclear receptor activity分子功能,condensed chromosome细胞过程。KEGG通路22条,主要涉及AGE-RAGE signaling pathway in diabetic complications。结论培土清心方治疗特应性皮炎是通过多成分、多基因、多通路,发挥抗炎、调节细胞免疫、抑菌的作用,这将为进一步研究此方作用机制提供参考,也为培土清心方治疗AD的基础、临床研究及新药研制提供一定的理论依据。
Objective To explore the molecular mechanism of PTQX formula in the treatment of atopic dermatitis based on network pharmacology and differential gene analysis of GEO chip.Methods The GEO database was searched to obtain the gene chip of atopic dermatitis,and the volcano map and heat map of differential genes were drawn.Using TCMSP,BATMAN-TCM and Drugbank databases,the active ingredients and genes of PTQX formula for atopic dermatitis were obtained,and cytoscape 3.7.2 was used to construct PTQX formula-atopic dermatitis-molecular gene network and topological network analysis.The R software was used to analyze the GO function and KEGG pathway enrichment of genes.Results 143 active ingredients of PTQX formula for atopic dermatitis were obtained.The main active ingredients for atopic dermatitis were quercetin,stigmasterol and luteolin.PTQX formula had 42 core genes corresponding to the disease,among which the important candidate genes were PPARG and so on.The important candidate genes for topological network analysis were TP53,NTRK1 and so on.GO analysis had a total of 516 biological pathways,8 cellular components,and 44 molecular functions,mainly involving the response to steroid hormone biological process,nuclear receptor activity molecular function,and condensed chromosome cellular processes.There were 22 KEGG pathways,mainly involving AGE-RAGE signaling pathway in diabetic complications.Conclusion PTQX formula treats atopic dermatitis through multiple components,multiple genes and multiple pathways,exerting anti-inflammatory,regulating cellular immunity and bacteriostatic effects,which will provide a reference for further research on the mechanism of this prescription.It also provides some theoretical basis for the basic,clinical research and new drug development of PTQX formula in the treatment of AD.
