F框WD40域蛋白7在胃癌组织的表达变化及对细胞增殖和上皮-间充质转化的影响 被引量:1

Expression of FBXW7 in gastric cancer and its effect on cell proliferation and epithelial-mesenchymal transition

摘要 目的观察F框WD40域蛋白7(FBXW7)在胃癌中的表达变化及对细胞增殖和上皮-间充质转化(EMT)的影响。方法选取2016年3月到2019年3月郑州大学附属医院南阳中心医院经手术切除的154例胃癌组织和癌旁组织作为研究材料,采用蛋白质印迹法(Western blot)分析肿瘤组织和癌旁组织中FBXW7蛋白表达水平。采用慢病毒在胃癌细胞SGC-7901中构建FBXW7过表达细胞株(FBXW7组)和对照细胞株(对照组)。采用5-乙炔基-2’脱氧尿嘧啶核苷(EdU)和细胞计数试剂盒(CCK-8)分析两组细胞增殖水平;采用Western blot分析两组细胞EMT标记分子蛋白表达水平。采用含基质胶的Transwell分析细胞的侵袭能力。组间比较采用t检验。结果与癌旁组织FBXW7蛋白表达水平(1.32±0.23)比较,肿瘤组织中FBXW7表达水平(0.52±0.12)显著下调,差异有统计学意义(t=3.091,P<0.05)。与对照组(1.98±0.27)比较,FBXW7组细胞72 h增殖程度(1.21±0.18)显著下降,差异有统计学意义(t=2.615,P<0.05)。与对照组[(54.32±7.32)%]比较,FBXW7组细胞EdU阳性率[(20.98±4.89)%]显著下降,差异有统计学意义(t=3.111,P<0.05)。与对照组细胞E-钙黏蛋白(E-cadherin)、紧密连接蛋白-1(ZO-1)、N-钙黏蛋白(N-cadherin)和波形蛋白(Vimentin)蛋白表达水平(0.89±0.19、1.20±0.28、1.39±0.21、1.51±0.26)比较,FBXW7组细胞E-cadherin和ZO-1表达水平(1.98±0.27、2.59±0.31)显著增加,而N-cadherin和Vimentin蛋白表达水平(0.40±0.11、0.53±0.19)显著下调,差异均有统计学意义(t=2.094、2.812、2.458、2.931,P<0.05)。与对照组细胞侵袭数量[(152.69±20.14)个]比较,FBXW7组细胞侵袭数量[(81.44±12.10)个]明显下降,差异有统计学意义(t=4.259,P<0.05)。结论FBXW7在胃癌组织中呈低表达,参与了胃癌细胞的增殖和EMT。 Objective To observe the expression of FBXW7 in gastric cancer and its effect on cell proliferation and epithelial-mesenchymal transition(EMT).Methods From March 2016 to March 2019,154 cases of gastric cancer tissues and adjacent tissues surgically removed in our hospital were collected as the study objects,and the expression levels of FBXW7 protein were analyzed by Western blotting.The overexpression cell line(FBXW7 group)and control cell line(control group)of FBXW7 were constructed by lentivirus in SGC-7901.Cell proliferation was analyzed by cell counting kit-8(CCK-8)and 5-Ethynyl-2’-deoxyuridine(EdU)staining.The protein expression level of EMT marker was analyzed by Western blotting.The invasion of two groups was analyzed by Transwell with matrix glue.Results The expression level of FBXW7 in tumor tissues(0.52±0.12)was significantly lower than that of the adjacent tissues(1.32±0.23,t=3.091,P<0.05).Compared with the control group(1.98±0.27),the proliferation of cells in FBXW7 group at 72 h(1.21±0.18)significantly decreased(t=2.615,P<0.05).Compared with the control group[(54.32±7.32)%],the positive rate of EdU in FBXW7 group[(20.98±4.89)%]significantly decreased(t=3.111,P<0.05).Compared with the control group(0.89±0.19,1.20±0.28,1.39±0.21,1.51±0.26),the expression levels of E-cadherin and ZO-1 in the FBXW7 group(1.98±0.27 and 2.59±0.31)significantly enhanced,and those of N-cadherin and Vimentin(0.40±0.11 and 0.53±0.19)significantly decreased(t=2.094,2.812,2.458,2.931,P<0.05).Compared with the control group[(152.69±20.14)cells],the number of invasive cells in FBXW7 group[(81.44±12.10)cells]significantly decreased(t=4.259,P<0.05).Conclusion The expression of FBXW7 is low in gastric cancer,which is involved in the proliferation of gastric cancer cells and EMT.
