摘要
于2015年7—8月份,对中国天山沿线地区(北纬39°25′12″~45°5′3″、东经75°2′28″~94°27′51″)的荒漠带、山地草原带、山地森林带、亚高山草原带、亚高山草甸带、高山草甸带的优势种和建群种进行了补充采集,每个物种尽可能选择2个或2个以上的个体,共采集113种250份植物样品;应用最大似然树(ML Tree)的方法,对天山沿线地区(中国)的306种植物进行DNA条形码分析,从物种与属水平统计了4个候选DNA条形码(ITS、matK、rbcL、trnH-psbA)在整体与多种属两方面的识别率差异,遴选天山沿线地区常见种子植物的DNA条形码。结果表明:整体上,ITS在单条形码中的识别率最高,为81.70%(种)、97.07%(属);多条形码组合分析中,ITS+matK+trnH-psbA的识别率最高,为86.66%(种)、96.59%(属),其次为ITS+matK(86.27%(种)、96.59%(属))、ITS+rbcL(86.66%(种)、95.12%(属))。对多种属的分析显示,ITS的识别率在单条形码中依然最高(69.28%(种)、98.08%(属));多条形码组合分析中,ITS+matK+trnH-psbA、ITS+matK的识别率最高,均为75.16%(种)、96.15%(属)。综合分析结果,依据通用性、识别性、经济性的原则,推荐ITS+matK为天山沿线地区常见种子植物的DNA条形码。
From July to August in 2015,250 samples of group and dominant plants(including 113 species)were collected for supplementary along Tianshan Mountain in China,between 39°25′12″N-45°5′3″N and 75°2′28″E-94°27′51″E.And each species were collected two or more individuals as much as possible.Four common single markers(ITS,matK,rbcL and trnH-psbA)were evaluated using 306 species along the Tianshan Mountains from two aspects with tree-based method(ML tree).In general,the resolution ratio of ITS was the highest(81.70%of species,97.07%of genus)in single,ITS+matK+trnH-psbA was the highest(86.66%of species,96.59%of genus)in combinations,followed by ITS+matK(86.27%of species,96.59%of genus)and ITS+rbcL(86.66%of species,95.12%of genus).The analysis of these genera containing more than one species revealed that ITS was the highest in single(69.28%of species,98.08%of genus)and ITS+maK+trnH-psbA and ITS+maK showed the highest resolution ratio(75.16%of species,96.15%of genus)in combinations.From the principles of universality,identification and economy,we suggest that the combination of ITS+matK should be regarded as the DNA barcodes for the common plants in Tianshan Mountains.
