知母皂苷AⅢ促进曲美替尼对肺癌A549细胞抑制活性的研究

Timosaponin AⅢEnchances Inhibition Effect of Trametinib on Lung Cancer A549 Cells

目的:观察曲美替尼与知母皂苷AⅢ联合作用时对肺癌A549细胞的抑制效应。方法:曲美替尼单独或与知母皂苷AⅢ联合作用于肺癌A549细胞;利用CCK8试剂盒检测细胞生长,单细胞集落形成实验检测细胞增殖能力,划痕实验检测细胞迁移能力,流式细胞术检测细胞周期分布。结果:曲美替尼与知母皂苷AⅢ联合使用时对A549细胞生长和迁移的抑制效果明显高于曲美替尼单独使用时的抑制效果(P<0.05);当曲美替尼与知母皂苷AⅢ联用时单细胞未能形成集落克隆;曲美替尼与知母皂苷AⅢ均能使A549周期阻滞于G0/G1期,但二者联合使用时的阻滞效果并未显著高于曲美替尼单独使用时的阻滞效果(P>0.05)。结论:知母皂苷AⅢ显著提高了曲美替尼对肺癌A549细胞生长、增殖和迁移能力的抑制。

Objective To investigate the inhibition effect of trametinib on lung cancer A549 cells when used combined with timosaponin AⅢ.Methods Trametinib was used only or combined with timosaponin A Ⅲ on A549 cells.CCK8 reagent was used to test growth of cells and colony formation from single cell was used to test proliferation capacity of cells.Wound Healing was used to test migrating ability of cells.Cell cycle was detected by flow cytometry.Results Compared with the cells treated with trametinib only,cells growth and migrating rate declined when cells treated with trametinib and timosaponin AⅢ(P<0.05).No colony grew out from single cell when cells treated with Timosaponin AⅢ and trametinib.Both Trametinib and timosaponin AⅢ blocked A549 cells at G0/G1 stage.The blocked proportion of cells treated with trametinib and timosaponin AⅢ combined was not markedly higher than which of cells treated with trametinib only(P>0.05).Conclusion Timosaponin AⅢ enhances the inhibition effect of trametinib on A549 cells through reducing growth,proliferation and migrating ability of cells.
