响应面法优化超声辅助提取金线莲总黄酮工艺研究 被引量:7

Optimization of Ultrasound-assisted Extraction of Total Flavonoids from Anoectochilus roxburghii(wall.)Lindl by Response Surface Method

摘要 采用响应面法优化设计化金线莲总黄酮超声波辅助提取工艺。以金线莲中总黄酮提取率为考察指标,根据中心组合(Box-Benhnken)试验设计原理,考察超声温度、超声时间、料液比以及乙醇浓度等影响因素对金线莲总黄酮提取效果的影响。最佳工艺为:提取温度45℃,超声时间58 min,料液比为1∶30,乙醇浓度为80%,预测总黄酮提取量为6.25 mg/g,验证实验实际提取量6.20 mg/g与之相比无显著差异。响应面分析法优化后的金线莲提取总黄酮工艺稳定可行,操作简便,提取效率高,可为金线莲总黄酮进一步的药理活性研究、制剂研制等提供基础。 The ultrasonic-assisted extraction process of total flavonoids from Anoectochilus roxburghii(wall.)Lindl by response surface method(RSM)was optimized.Taking the extraction rate of total flavonoids from Anoectochilus roxburghii as the index,according to the design principle of central combination(Box-Benhnken),the effects of ultrasonic temperature,ultrasonic time,ratio of material to liquid and ethanol concentration on the extraction of total flavonoids from Anoectochilus roxburghii were investigated.The optimum conditions were as follows:extraction temperature was 45℃,ultrasonic time was 58 min,ratio of material to liquid was 1:30,ethanol concentration was 80%,and the predicted total flavonoid extraction amount was 6.25 mg/g.It was verified that there was no significant difference in the actual extraction amount of 6.20 mg/g.The optimization of the response surface analysis method for extracting total flavonoids from Anoectochilus is stable and feasible,easy to operate,and high in extraction efficiency.It can provide a basis for further pharmacological activity research and preparation development of the total flavonoids of Anoectochilus.
