摘要
采用数码显微摄像仪鉴定新疆芍药粉末的显微特征;采用Folin-Ciocalteu比色法测定新疆芍药中总多酚的含量;采用连续稀释法检测新疆芍药总多酚对变异链球菌的体外抑菌活性。结果表明,新疆芍药粉末的显微特征较为明显,可见纤维、非腺毛、螺纹导管、气孔、栅栏细胞;标准曲线的线性回归方程为y=137.25x+0.09221(R^2=0.999),线性范围为0.001~0.005 mg·mL^-1,平均加标回收率为99.84%,RSD为2.27%;新疆芍药总多酚含量为(9.58±1.03)mg·g^-1,其对变异链球菌的最低抑菌浓度为72.24 mg·mL^-1。Folin-Ciocalteu比色法简便、准确、稳定,可作为测定新疆芍药中总多酚含量的可靠方法。新疆芍药总多酚具有抑菌作用,可作为潜在的抑菌活性成分,为有效开发利用新疆芍药资源、初步筛选安全有效的防龋药物奠定了基础。
We identified the microscopic characteristics of P.sinjiangensis K.Y.Pan powder by digital microcamera,determined the content of total polyphenols from P.sinjiangensis K.Y.Pan by Folin-Ciocalteu colorimetry,and detected the in vitro antibacterial activity of total polyphenols from P.sinjiangensis K.Y.Pan to Streptococcus mutans by a continuous dilution method.The results show that the microscopic characteristics of P.sinjiangensis K.Y.Pan powder are obvious,such as fibers,non-glandular hairs,threaded ducts,stomata,and palisade cells.The linear regression equation of the standard curve is y=137.25x+0.09221(R^2=0.999),the linear range is 0.001-0.005 mg·mL^-1,the average adding standard recovery is 99.84%,and the RSD is 2.27%.The content of total polyphenols from P.sinjiangensis K.Y.Pan is(9.58±1.03)mg·g^-1,whose MIC to Streptococcus mutans is 72.24 mg·mL^-1.Folin-Ciocalteu colorimetry is simple,accurate,and stable,which can be used as the reliable method to determine the content of total polyphenols from P.sinjiangensis K.Y.Pan.The total polyphenols from P.sinjiangensis K.Y.Pan have antibacterial effect and can be used as potential antibacterial active ingredients,which lays a foundation for the effective development and utilization of P.sinjiangensis K.Y.Pan and the preliminary screening of safe and effective anti-carious drugs.
