白花九里明提取物中3种有机酸及总黄酮、总有机酸的含量测定 被引量:3

摘要 建立了白花九里明提取物中原儿茶酸、绿原酸、咖啡酸、总黄酮和总有机酸含量的测定方法。采用双波长切换高效液相色谱法测定原儿茶酸、绿原酸、咖啡酸的含量,采用酶标法,分别以槲皮素、原儿茶酸为对照品,在373 nm、292 nm下测定总黄酮和总有机酸含量。结果表明,原儿茶酸、绿原酸、咖啡酸分别在0.348~3.132μg、0.4~3.6μg、0.142~1.278μg范围内与峰面积呈良好的线性关系(R^2=0.9999),平均加标回收率分别为98.07%、97.49%、99.81%,RSD分别为1.35%、1.42%、0.43%;槲皮素、原儿茶酸分别在8.24~41.20μg·mL^-1、10.8~54.0μg·mL^-1浓度范围内与吸光度呈良好的线性关系,R^2分别为0.9998和0.9999,平均加标回收率分别为102.4%、101.1%,RSD分别为0.75%、1.07%。白花九里明提取物中原儿茶酸、绿原酸、咖啡酸、总黄酮和总有机酸的含量分别为1.29 mg·g^-1、3.68 mg·g^-1、0.81 mg·g^-1、22.33 mg·g^-1和190.92 mg·g^-1。该方法简便、准确、重复性好,可作为白花九里明提取物中原儿茶酸、绿原酸、咖啡酸、总黄酮和总有机酸的含量测定方法。 We established a determination method for protocatechuic acid,chlorogenic acid,caffeic acid,total flavonoids,and total organic acids in Blumea megacephala extract.Moreover,we determined the contents of protocatechuic acid,chlorogenic acid,and caffeic acid by dual wavelength switching high performance liquid chromatography.Furthermore,we determined the contents of total flavonoids and total organic acids at 373 nm and 292 nm with quercetin and protocatechuic acid as the control,respectively.The results show that protocatechuic acid,chlorogenic acid,and caffeic acid show a good linear relationship(R^2=0.9999)with peak areas in the range of 0.348-3.132μg,0.4-3.6μg,0.142-1.278μg,respectively,the average adding standard recoveries are 98.07%,97.49%,and 99.81%,respectively,and the RSD are 1.35%,1.42%,and 0.43%,respectively.Total flavonoids and total organic acids show a good linear relationship with absorbance in the range of 8.24-41.20μg·mL^-1 and 10.8-54.0μg·mL^-1,respectively,the correlation coefficient R^2 are 0.9998 and 0.9999,respectively,the average adding standard recoveries are 102.4%and 101.1%,respectively,and the RSD are 0.75%and 1.07%,respectively.The contents of protocatechuic acid,chlorogenic acid,caffeic acid,total flavonoids,and total organic acids in Blumea megacephala extract are 1.29 mg·g^-1,3.68 mg·g^-1,0.81 mg·g^-1,22.33 mg·g^-1,and 190.92 mg·g^-1,respectively.This method is simple,accurate,and reproducible,and can be used for the determination of protocatechuic acid,chlorogenic acid,caffeic acid,total flavonoids,and total organic acids in Blumea megacephala extract.
