摘要
目的观察温阳振衰颗粒对慢性心力衰竭(CHF)模型大鼠心肌LncRNA BIC/miR-155调控P38MAPK表达的影响。方法健康SD大鼠40只,随机抽取10只为正常对照组,余30只建立CHF模型后随机分为模型对照组、温阳振衰组和阳性对照组,各组连续干预4 w。检测各组心肌LncRNA BIC、miR-155及P38MAPK的表达水平。结果与正常对照组相比,各实验组LncRNA BIC和miR-155表达明显下调(P<0.05),温阳振衰组较模型对照组和阳性对照组显著下调缓慢(P<0.05)。与正常对照组相比,各实验组p-P38MAPK表达明显上调(P<0.05),温阳振衰组较模型对照组和阳性对照组显著上调缓慢(P<0.05)。结论温阳振衰颗粒治疗CHF的机制可能是对心肌组织中的LncRNA BIC和miR-155进行调控进而抑制P38MAPK的表达。
Objective To elucidate the effects of Wenyangzhenshuai granule on LncRNA BIC/miR-155 regulating P38 MAPK expressions in chronic heart failure rats.Methods 10 SD rats were randomly extracted from 40 rats into normal control group and the remaining 30 rats were used as models and randomly divided into model control, Wenyangzhenshuai and positive control groups, with each group 10 rats. Each group was given corresponding treatment for 4 weeks. LncRNA BIC, miR-155 and P38 MAPK expressions in these groups were detected by Western blot and RT-PCR.Results LncRNA BIC and miR-155 in all the groups were lower than those in normal control group(P<0.05), those in Wenyangzhenshuai group were higher than those of model and positive groups(P<0.05). p-P38 MAPK in all the groups was higher than that in normal control group(P<0.05), that in Wenyangzhenshuai group was lower than that of model and positive groups(P<0.05).Conclusions Wenyang zhenshuai granule could enhance LncRNA BIC and miR-155 and inhibit the activation of P38 MAPK, which might be one of the important mechanisms for treatment of chronic heart failure.
