右美托咪定对重度烧伤SD大鼠肝损伤的影响 被引量:2

The effects of Dexmedetomidine on liver injury in severely burned SD rats

摘要 目的评价右美托咪定对重度烧伤SD大鼠肝损伤的影响。方法将30只SD大鼠随机分为空白对照组(C组)、烧伤组(B组)和右美托咪定组(D组)。建立大鼠30%体表面积烧伤模型,后按Parkland公式补液抗休克治疗。D组烧伤后即刻腹腔注射右美托咪定30μg/kg,B组腹腔注射等容量生理盐水。于烧伤后24h时取血样,测定血清中谷丙转氨酶(AST)、谷草转氨酶(ALT)的含量,然后处死大鼠取其肝组织,HE染色检测肝组织的病理学变化。ELISA法检测肝组织的肿瘤坏死因子α(TNF-α)水平。TUNEL染色检测肝细胞凋亡率。结果与C组比较,B组和D组AST、ALT明显升高,肝组织中TNF-α含量增多,肝细胞凋亡指数升高(P<0.05)。与B组比较,D组AST、ALT明显降低,肝组织中TNF-α含量下降,肝细胞凋亡指数降低(P<0.05)。B组肝脏病理学损伤明显,D组肝脏病理学损伤明显减轻。结论右美托咪定可减轻重度烧伤SD大鼠肝脏炎症因子的生成和肝细胞凋亡,具有一定能的肝保护作用。 Objective To evaluate the effect of dexmedetomidine on liver injury in severely burned SD rats.Methods 30 SD rats were randomly divided into control group(group C),burn group(group B)and Dexmedetomidine group(group D).30%of the total body surface was shaved and exposed to 94℃water for 12 seconds.Immediately after burn,the rats were resuscitated with isotonic saline according to Parkland formula.Sham burn Was produced in C group.In group D,the rats received inraperitoneal injection of Dexmedetomidine 30μg/kg immediately after burn,and the equal volume of normal saline was injected in group B.Blood samples were collected at 24 hours after burn to determinethe levels of AST and ALT.Then killed all rats and removed to observe the pathological changes of liver tissues by HE staining,the levels of TNF-a,by ELISA;the apoptotic rate of liver cells was detected by TUNEL method.Results Compared with group C,AST and ALT in group B and D increased significantly,TNF-a in liver tissue increased,and hepatocyte apoptosis index increased(P<0.05).Compared with group B,AST and ALT in group D decreased significantly,TNF-a in liver tissue decreased.and hepatocyte apoptosis index decreased(P<0.05).The pathological damage of liver in group B were obvious,in group D was significantly reduced.Conclusion Dextrometridine has protective effect on liver injury in severely burned SD rats by reducing the production of inflammatory factors and hepatocyte apoptosis.
