摘要
目的制备针对严重急性呼吸系统综合症冠状病毒2(SARS-CoV-2)的单克隆抗体,用于抗原诊断。方法收集分泌抗SARS-CoV单克隆抗体的杂交瘤细胞上清液,并用Western blot和间接酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)检测单克隆抗体与SARS-CoV-2 N蛋白之间的反应。通过免疫荧光,检测单克隆抗体与Vero细胞培养物中SARS-CoV-2的反应性。结果从杂交瘤细胞中获得了两种单克隆抗体,分别称为A1和A2。它们都可以与SARS-CoV-2 N蛋白发生反应,而A2也可以识别SARS-CoV N蛋白。这些单克隆抗体可用于ELISA和免疫荧光实验。结论获得了2种针对SARS-CoV-2 N蛋白的单克隆抗体,为发展SARS-CoV-2抗原检测方法提供了基础。
Objective To prepare monoclonal antibodies against Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2)for developing antigen diagnostics.Methods The supernatants of hybridoma cell lines secreting monoclonal antibodies against SARS-CoV were collected and were subjected to examinate the reaction between the monoclonal antibodies and SARS-CoV-2 N protein by using western blot and indirect enzyme-linked immunosorbent assay(ELISA).The reactive monoclonal antibodies were used to detect SARS-CoV-2 in Vero cell cultures by using immunofluorescence assay.Results Two monoclonal antibodies,termed A1 and A2,were obtained from the hybridoma cell lines.They can both react with SARS-CoV-2 N protein,while A2 can also recognize SARS-CoV N protein.These monoclonal antibodies can be used in ELISA and immunofluorescence experiments.Conclusions Two monoclonal antibodies against SARS-CoV-2 N protein were obtained,providing materials for future development of SARS-CoV-2 antigen detection.
