摘要
目的观察胃癌组织长链非编码RNA X染色体失活特异转录本(lncRNA XIST)、微小RNA-337-3p(miR-337-3p)表达变化,并分析二者表达的关系及其与患者临床病理特征的关系。方法选择胃癌患者79例,取活检或手术切除的胃癌组织及其配对的癌旁正常组织,采用RT-qPCR法检测lncRNA XIST、miR-337-3p表达。比较胃癌组织与癌旁正常组织lncRNA XIST、miR-337-3p表达变化,并分析胃癌组织lncRNA XIST、miR-337-3p表达的关系及其与患者临床病理特征的关系。结果胃癌组织lncRNA XIST相对表达量高于癌旁正常组织,miR-337-3p相对表达量低于癌旁正常组织(t分别为19.081、13.459,P均<0.05)。Pearson线性相关分析显示,胃癌组织lncRNA XIST相对表达量与miR-337-3p相对表达量呈负相关关系(r=-0.672,P<0.05)。胃癌组织lncRNA XIST、miR-337-3p相对表达量均与TNM分期、组织分化程度、淋巴结转移有关(P均<0.05),与性别、年龄、肿瘤直径无关(P均>0.05)。结论胃癌组织lncRNA XIST高表达、miR-337-3p低表达,二者表达呈负相关关系,并且二者共同参与胃癌的发生、发展。lncRNA XIST、miR-337-3p有望成为胃癌早期诊断和靶向治疗的分子生物标志物。
Objective To observe the expression changes of long non-coding RNA X inactive-specific transcript(lncRNA XIST)and microRNA-337-3p(miR-337-3p)in the gastric cancer tissues,and to analyze the relationship between their expression and the clinicopathological characteristics of patients.Methods Seventy-nine patients with gastric cancer were selected.The gastric cancer tissues resected by biopsy or surgery and their paired adjacent normal tissues were taken,and the expression levels of lncRNA XIST and miR-337-3p were detected by RT-qPCR.The expression changes of lncRNA XIST and miR-337-3p in the gastric cancer tissues and their paired adjacent normal tissues were compared,and the relationship between the expression of lncRNA XIST and miR-337-3p in the gastric cancer tissues and their clinicopathological characteristics were analyzed.Results The relative expression of lncRNA XIST in the gastric cancer tissues was higher than that in the adjacent normal tissues,and the relative expression of miR-337-3p was lower than that in the adjacent normal tissues(t=19.081 and 13.459,respectively,both P<0.05).Pearson linear correlation analysis showed that the relative expression of lncRNA XIST in the gastric cancer tissues was negatively correlated with the relative expression of miR-337-3p(r=-0.672,P<0.05).The relative expression levels of lncRNA XIST and miR-337-3p in the gastric cancer tissues were related to TNM stage,tissue differentiation degree,and lymph node metastasis(all P<0.05),but not to gender,age or tumor diameter(all P>0.05).Conclusions The high expression of lncRNA XIST and the low expression of miR-337-3p in gastric cancer tissues are related to the occurrence and development of gastric cancer.LncRNA XIST and miR-337-3p are expected to be molecular biomarkers for early diagnosis and targeted therapy of gastric cancer.
