温敏型脂质纳米粒的制备及其识别微波消融边界效果 被引量:1

摘要 目的制备包裹液态全氟化碳的温敏型脂质纳米粒(HLNP),观察其体外热相变效果及体外识别微波消融边界效果。方法制备包裹液态全氟化碳的HLNP,利用光镜实时观察其在加热板作用下的相变情况,以超声诊断仪实时观察微波加热含HLNP凝胶孔洞模型的显像效果。在离体牛肝微波消融范围周边注入HLNP,实时观察超声回声变化及消融后牛肝大体变化。结果所制备HLNP平均粒径(507.90±101.70)nm;光镜下HLNP在加热板温度为45℃时开始出现液气相转变,达50℃后产生大量微气泡;微波热消融作用下,含HLNP的凝胶模型超声回声明显增强。离体牛肝实验显示,在微波消融气化区周边注入HLNP后超声回声明显增强,组织从红色变为白色。结论所制备的HLNP能在加热和微波热消融作用下发生液气相转变,有望成为可实时监控微波消融效果,并评估微波消融边界的热敏剂。 Objective To prepare heat-sensitive lipid nanoparticles(HLNP)capsuling liquid perfluorocarbon,and to observe the phase-shift of HLNP under heating and the effect in microwave ablation in vitro.Methods HLNP were prepared,and the real-time phase-shift effect of HLNP caused by heating plate was observed using the light microscope,and HLNP-included void hydrogel phantom heated by microwave on ultrasound imaging was observed.Then the echo changes of ultrasound and macroscopic changes of bovine liver injected with HLNP around the microwave ablation range were explored.Results The mean particle size of the prepared HLNP was(507.90±101.70)nm.Phase-shift of HLNP began when the temperature of heating plate reached 45℃,and a large number of microbubbles were gradually generated under 50℃.The ultrasonic echo of HLNP significantly enhanced after microwave heating.In vitro bovine liver experiment showed that the bovine liver injected with HLNP changed from red to white after microwave ablation and ultrasonic echo significantly enhanced around the microwave ablation gasification zone.Conclusion Liquid-gas phase transition of the prepared HLNP happened under heating and microwave thermal ablation,indicating that the prepared HLNP may be used as a kind of thermal sensitizer for identifying microwave ablation boundary and real-time monitoring effect for microwave ablation.
