摘要
目的检测靶向纳米粒造影剂在VX2兔舌癌MLNs中的双模态显影以及初步研究其对肿瘤的作用。方法采用双乳化法制备多功能相变纳米粒。建立兔舌癌转移淋巴结模型,证实该造影剂可增强体内MLNs显影,寻找显影规律。随机分五组VX2兔为实验对象,注射纳米粒2 ml,以激光激发,获取注射前后不同时间点的光声图像及信号值。VX2兔注射造影剂2d/次,持续1个月,观察其生存情况以及肿瘤生长状况,结束后行淋巴结及重要器官病理检查。结果成功建立动物模型。VX2兔注射造影剂后15min开始显影,至4h达峰值;靶向微泡组光声显影效果在注射后各时间点均明显强于非靶向微泡组(P<0.05)。在观察期内,存活VX2兔淋巴结较小,凋亡实验显示肿瘤细胞明显凋亡,重要脏器未见明显异常。结论该新型纳米粒造影剂在VX2兔MLNs中具有良好的双模态显影能力。并且该造影剂可作用于MLNs,延长VX2兔的生存时间,有望成为新型口腔鳞癌MLNs的治疗方式。
%Objective To discover the imaging bimodal development of targeted nanoparticle contrast agents in VX2 rabbit tongue cancer MLNs was examined and the effects on tumors were preliminarily studied.Methods Multifunctional phase change nanoparticles were prepared by double emulsion method.The rabbit tongue cancer for MLNs model was established,which confirmed that the contrast agent can enhance the development of MLNs in the body and search for developmental rules.Five groups of VX2 rabbits were randomly divided into experimental subjects.2 ml of nanoparticles were injected and laser-excited to obtain photoacoustic images and signal values at different time points before and after injection.VX2 rabbits were injected with contrast agent for 2 days/time for 1 month.The survival and tumor growth status were observed.After the end,lymph nodes and pathological examination of vital organs were performed.Results Animal models were successfully established.Imaging to develop after 15 minutes of contrast injection,peaking at 4h;the photoacoustic development effect of the targeted microbubble group was significantly stronger than that of the non-targeted microbubble group at each time point after injection(P<0.05).During the observation period,the viable VX2 rabbit lymph nodes were small,and the apoptosis experiment showed that the tumor cells were obviously apoptotic,and no obvious abnormalities were observed in the important organs.Conclusion The novel nanoparticle contrast agent has good bimodal development ability in VX2 rabbit MLNs.Moreover,the contrast agent can prolong survival time of VX2 rabbits and act on MLNs,which is expected to be the new treatment for oral squamous cell carcinoma MLNs.
