摘要
探讨香兰素通过NF-κB信号通路抑制巨噬细胞活化及分化的相关机制,以明确香兰素抑炎抗炎的相关途径,为更加合理的使用香兰素提供理论依据。以巨噬细胞为研究对象,分为对照组、模型组和香兰素处理组。模型组使用LPS处理巨噬细胞以构建巨噬细胞活化模型,香兰素处理组为使用LPS处理后添加香兰素,对照组不予处理;使用qRT-PCR,流式细胞仪检测,western blot以及免疫荧光等方法对香兰素抑制巨噬细胞活化的机制进行探讨。结果显示,在使用LPS处理后,巨噬细胞内IL-1βmRNA表达量显著升高、巨噬细胞活化标记物活性显著增强、p65亚基的磷酸化水平显著上调并且其入核显著增强。而在使用香兰素处理后,上述现象均遭到明显抑制。综合以上结果分析得出结论,香兰素是通过下调p65亚基的活性阻碍其活化入核从而抑制巨噬细胞活化,达到其抗炎抑炎的目的。
To provide a theoretical basis for more rational use of vanillin,investigate the mechanism of vanillin inhibiting macrophage activation and differentiation through NF-κB signaling pathway,the related pathways of vanillin anti-inflammatory and anti-inflammatory were clarified.Macrophages were used as research objects and divided into control group,model group and vanillin treatment group.In the model group,macrophages were treated with LPS to build a macrophage activation model.Vanillin was added to the vanillin group after treatment with LPS,while no treatment was given to the control group.qRT-PCR,flow cytometry,western blot and immunofluorescence were used to investigate the mechanism of vanillin inhibiting macrophage activation.The results showed that,after LPS treatment,the expression of IL-1βmRNA in macrophages was significantly increased,the activity of macrophage activation markers was significantly enhanced,the phosphorylation level of p65 subunit was significantly up-regulated and the amount of nuclear entry was significantly increased.After the treatment with vanillin,the above phenomenon was significantly inhibited.Based on the analysis,it is concluded that vanillin inhibited the activation of macrophages by down-regulating the activity of p65 subunit and prevented its activation into the nucleus,thus achieved the purpose of anti-inflammatory and anti-inflammatory.
