Expression and significance of GLDN gene in colorectal cancer

目的探讨GLDN基因在结直肠癌中的表达及其意义。方法从GEO数据库下载GSE50760数据包。利用R语言对数据包进行分析,得出与结直肠癌相关的差异表达基因,并绘制热图和火山图。运用starbase在线数据库对获得的差异基因在结直肠癌与癌旁组织中的表达水平进行分析。进一步利用OncoLnc在线数据库进行差异基因与结直肠癌的预后分析。在临床样本层面,利用qPCR及免疫组化技术探索GLDN基因在结直肠癌中的表达差异。结果通过分析GSE50760数据包获得前100个差异表达的基因。利用starbase在线数据库对这100个差异表达基因的表达水平进行分析发现,GLDN基因在结直肠癌组织中的表达比癌旁组织明显下调。生存分析也提示,当GLDN基因高表达时,结直肠癌患者的预后相对较好。qPCR及免疫组化技术也验证了GLDN临床样本的癌旁组织中高表达,并且GLDN表达量与患者预后密切相关。结论GLDN是结直肠癌的潜在抑癌基因,并且可以作为判断结直肠癌患者预后的重要生物学标记物。

Objective To investigate the expression of GLDN gene in colorectal cancer and its significance.Methods Download the GSE50760 data package from the GEO database.Using R language to analyze the data packets,the differentially expressed genes related to colorectal cancer were obtained,heat maps and volcano maps were drawn.The starbase online database was used to analyze the expression levels of the obtained differential genes in colorectal cancer and adjacent tissues.The oncoLnc online database was used to analyze the prognosis of differential genes and colorectal cancer.At the level of clinical samples,qPCR and immunohistochemical techniques were used to explore the expression differences of GLDN in colorectal cancer.Results The top 100 differentially expressed genes were obtained by analyzing the GSE50760 data package.Using the starbase online database to analyze the expression levels of these 100 differentially expressed genes,it was found that the expression of GLDN gene in colorectal cancer tissues was significantly down-regulated than that in adjacent tissues.Survival analysis also suggests that when the GLDN gene was highly expressed,the prognosis of colorectal cancer patients was relatively good.qPCR and immunohistochemical techniques had also confirmed that GLDN clinical samples were highly expressed in paracancerous tissues,and the expression of GLDN was closely related to the prognosis of patients.Conclusion GLDN is a potential tumor suppressor gene for colorectal cancer,and it can be used as an important biomarker for the prognosis of colorectal cancer patients.
