NF-κB炎症信号通路相关基因SNPs 与胃癌预后的相关性研究

Correlation study between NF-κB inflammatory signaling pathway related gene SNPs and prognosis of gastric cancer

目的探索NF-κB炎症信号通路相关基因SNPs与胃癌预后的相关性,寻找影响胃癌预后的分子生物标志物。方法收集2013年4月至2014年3月在仙游县医院经病理确诊的167例胃癌患者血液,提取血标本基因组DNA,进行SNPs检测;通过病案资料整理和面对面访谈方式获得患者的疾病信息和预后状况,运用生存分析方法剖析NF-κB炎症信号通路相关基因SNPs与胃癌预后的相关性。结果多因素分析显示rs3755867位点为独立的影响因素;按年龄和肿瘤分期(TNM)联合分层,结果显示IL-1B的rs1143627位点、NFKB1的rs3755867位点、RELA的rs7119750位点的多态性与胃癌预后相关;基因多态性位点联合作用分析,同时携带rs1143627位点、rs3755867位点不良基因型的胃癌患者预后不良的风险大,HR为1.92(95%CI=1.04~3.52)。结论NF-κB炎症信号通路相关基因多个单核苷酸位点与胃癌的预后有关。

Objective To explore the correlation between NF-κB inflammatory signaling pathway related gene SNPs and the prognosis of gastric cancer, and to find molecular biomarkers affecting the prognosis of the cancer. Methods The blood of 167 cases of gastric cancer diagnosed pathologically in Xianyou County Hospital was collected from Apr. 2013 to Mar.2014, and the SNPs were detected in genomic DNA of blood samples. The patient’s disease information and prognosis were obtained through medical record data and face-to-face interview, the correlation between NF-κB inflammatory signaling pathway related gene SNPs and the prognosis of gastric cancer were analyzed by survival analysis method. Results Multivariate analysis showed that rs3755867 locus was an independent influencing factor. Combined stratification by age and tumor stage(TNM), the results showed that the polymorphism in rs1143627 locus of IL-1 B gene, rs3755867 locus of NFKB1 gene and rs7119759 locus of RELA gene were associated with the prognosis of gastric cancer.Join effect analysis of gene polymorphism loci showed that there was a high risk of poor prognosis in patients with gastric cancer with poor genotype of rs1143627 locus and rs3755867 locus, HR was 1.92(95% CI =1.04~3.52). Conclusion Multiple single nucleotide sites of NF-κB inflammatory signaling pathway related genes were correlated to the prognosis of gastric cancer.
