基于网络药理学探讨左归丸维持间充质干细胞干性的分子机制 被引量:3

Molecular Mechanism of Zuogui Pill on Maintaining the Stemness of Mesenchymal Stem Cells Based on Network Pharmacology

摘要 目的:采用网络药理学的方法,探讨左归丸维持间充质干细胞干性的分子机制,进一步探索左归丸药效成分的潜在作用靶点。方法:在TCMSP平台及BATMAN-TCM中检索左归丸的活性成分及基因靶点,在Genecards及人类孟德尔遗传在线数据库检索并获取间充质干细胞干性相关基因,应用Cytoscape3.7.2软件构建左归丸-间充质干细胞干性-靶基因网络图,并进一步生成蛋白质-蛋白质互作网络,寻找网络关键基因。最后利用R3.6.0软件,进行GO功能注释和KEGG通路富集分析。结果:本研究中共筛选出左归丸活性成分105种,其中有对应靶点的活性成分64种;获得间充质干细胞干性相关靶点17878个,与左归丸活性成分作用靶基因取交集后得到共同靶基因317个,主要富集于175个生物过程和137条信号通路上。结论:左归丸可能通过影响细胞增殖、分化、凋亡、抑制炎症反应、抗病毒、影响相关蛋白质及酶的合成,从而维持间充质干细胞干性、延缓衰老。 Objective:To study the mechanism of Zuogui Pill to maintain the stemness of mesenchymal stem cells by network pharmacology,and to explore its potential target.Methods:The active components and gene targets of Zuogui Pill were screened in TCMSP and BATMAN-TCM.Mesenchymal stem cells related genes were acquired in Genecards and human Mendelian Inheritance online database.The"components of Zuogui Pill-stemness of mesenchymal stem cells-target"network was constructed by Cytoscape 3.7.2.Furthermore,the protein-protein interaction network was generated to search for the key genes.The GO function annotation and KEGG signal pathway enrichment analysis were performed by software R3.6.0.Results:Totally 105 active components of Zuogui Pill were screened,involved 64 active components with corresponding targets.17878 targets related to the stemness of mesenchymal stem cells were screened,after crossing with the active component corresponding targets of Zuogui Pill,317 common targets were obtained,which mainly enriched in 175 biological processes and 137 signaling pathways.Conclusion:Zuogui Pill may maintain the stemness of MSCs and delay aging by regulating proliferation,differentiation and apoptosis of cells,inhibiting inflammatory reaction,playing antiviral function and affecting the synthesis of related proteins and enzymes.
