水稻种子特异表达OsEnS58基因的克隆与表达分析

摘要 水稻是世界重要的粮食作物之一,胚乳是水稻营养物质运输与储存的场所。为探究水稻胚乳特异表达基因Os EnS58的结构与功能,本研究以粳稻‘中花11’为材料,通过生物信息学和分子生物学分析该基因的结构与表达。生物信息学分析显示,OsEnS58基因全长766 bp,其启动子包含多种胚乳特异表达、光响应和激素调控相关的元件。qRT-PCR分析发现,OsEnS58在授粉后10 d种子中高表达;原位杂交进一步证实该基因在授粉后7 d和10 d种子的内种皮中高表达,这说明OsEnS58为胚乳特异表达基因。共表达分析显示,与OsEnS58共表达基因参与水稻转录调控、胁迫响应和转运调控。本研究结果表明OsEnS58基因在水稻种子发育,尤其是储藏物质的合成、转运与储藏过程中发挥作用,为进一步研究其在水稻中的分子机制提供了科学依据。 Rice is one of the most important cereal crop for word population,and its endosperm is the crucial place where nutrients are transported and stored.To investigate potential function of OsEnS58,we analyzed the structure and expression of the gene by bioinformatics analysis and molecular techniques in'Zhonghua11'.Bioinformatics analysis showed that the total length of OsEnS58 gene was 766 bp,and its promoter region contained various elements related to endosperm-specific expression,light response and hormone regulation.The results of qRT-PCR indicated that OsEnS58 was specifically expressed in 10-day-old seeds after pollination.It was con-firmed that the expression of OsEnS58 was specifically expressed in the endopleura of developing seed 7 d,and 10 d after pollination by in-situ hybridization.It indicated OsEnS58 was a typical endosperm-specific gene in rice.Further analysis revealed that OsEnS58 together with its co-expressing genes were involved in transcriptional regulation,stress response and transport regulation in rice.These results suggest that OsEnS58 plays a crucial role in rice seed development,especially in the synthesis,transport and storage of reserve substance,and provide sci-entific basis for further research of its molecular mechanism in rice.
