赤霉素诱导下珠芽魔芋顶芽激素含量的变化 被引量:3

Changes of Plant Hormone Contents in GA 3-induced Apical Bud of Amorphophallus bulbifer

摘要 魔芋产业是中西部地区农业产业结构调整、农民脱贫致富的重要支柱产业之一。外施赤霉素能够诱导魔芋顶芽的成花转变,是提高魔芋繁殖系数的有效手段。为探讨魔芋顶芽不同激素对外施赤霉素的响应,以珠芽魔芋(Amorphophallus bulbifer)为试验材料,利用超高效液相色谱法测定经GA3处理后,魔芋顶芽ZT、GA3、IAA和ABA含量的变化。结果表明,在选用0.6%乙酸和甲醇溶液作为流动相、梯度洗脱的条件下,四种激素分离效果较好,每个样品检测时间在10 min内。GA3处理后,魔芋顶芽组织内GA3含量显著上升,在处理后3~7 d GA3含量保持较高水平,约为对照的2.4~3.3倍。IAA含量在GA3处理后缓慢升高,在处理后14 d约为对照的2.5倍。ABA含量在GA3处理后降低,在处理后3 d未检测到ABA,在处理后7 d魔芋顶芽ABA含量约为对照的0.4倍。ZT含量在GA3处理前后无明显变化。综上,外施GA3处理下,魔芋顶芽组织内GA3和IAA含量升高,ABA含量降低,这些变化可能是GA3诱导下魔芋成花转变的影响因素之一。本研究通过分析GA3处理下魔芋顶芽激素的变化,为深入理解赤霉素调控魔芋成花的机理、指导珠芽魔芋快繁技术提供一定的理论依据。 Konjac industry is one of the most important pillar industry in the adjustment of agriculture structure,as well as in helping alleviate the poverty in the midwest of China.GA 3-induced floral transformation is an effec-tive method in increasing propagation coefficient in Amorphophallus.To study the changes of contents of plant hormones induced by GA 3 in A.%bulbifer,UPLC was used to determine the contents of four endogenous hormones:ZT,GA 3,IAA and ABA in terminal bud of A.%bulbifer after GA 3 treatment.Under the condition of 0.6%acetic acid and methanol solution as mobile phase and gradient elution,the four hormones was separated effectively in 10 minutes.The content of GA 3 in terminal bud was up-regulated by GA 3-treatment,and were 2.4 and 3.3 times of control 3 and 7 d after GA 3-treatment,respectively.The content of IAA was up-regulated by GA 3-treatment,and was 2.5 times of control 14 d after GA 3-treatment.To the contrary,the content of ABA was down-regulated by ex-ogenous GA 3.ABA was not detected 3 d after GA 3-treatment,and the content of ABA was 0.4 times of control 7 d after GA 3-treatment.The content of ZT showed no significate change after GA 3 treatment.Above all,GA 3-treat-ment up-regulated the contents of endogenous GA 3 and IAA,and down-regulated the contents of endogenous ABA of terminal bud of A.bulbifer,changes of the contents of plant hormones after GA 3-treatment might be one of the determinant of GA 3-induced flowering transition in A.bulbifer.This study demonstrated the changes of contents of plant hormones induced by GA 3 in A.%bulbifer,which provided theoretical basis for further study of GA 3-in-duced flowering transition in rapid propagation technique of A.%bulbifer.
