益气活血中药对心衰大鼠心肌PGC-1α能量代谢影响的实验研究 被引量:11

Experimental Study on Effect of Yiqi Huoxue Recipe on Energy Metabolism of Myocardial PGC-1α in Heart Failure Rats

摘要 目的观察益气活血中药对心梗后心衰(heart failure,HF)大鼠心肌能量代谢的影响。方法选用80只SPF级SD雄性大鼠,从中随机法选取20只作为空白对照组,剩余60只大鼠采用冠脉结扎法并配合力竭式游泳、节食法以制作心衰大鼠模型,将造模成功的大鼠随机分为西药组、模型组、益气活血中药组。给予空白对照组、模型组等容积蒸馏水日1次灌胃,西药组给予赖诺普利生药1.5 mg/(kg·d)剂量日1次灌胃,益气活血中药组予以益气活血中药10.0 g/(kg·d)日1次灌胃。持续给药28 d,大鼠禁食不禁水12 h后,取心肌组织及血浆:ELISA法检测BNP浓度,免疫蛋白印迹法、实时荧光定量PCR和检测PGC-1α基因和蛋白的表达。结果与空白对照组相比较,益气活血中药组、模型组、西药组PGC-1αmRNA及蛋白表达均降低,BNP浓度均升高,有统计学差异(P<0.05)。与模型组比较,西药组、益气活血中药组PGC-1αmRNA及蛋白表达均升高,BNP浓度降低,均有统计学意义差异(P<0.05);西药组与益气活血中药组两者比较无统计学差异(P>0.05)。结论益气活血中药可通过激活PGC-1α因子,增加心肌能量代谢,增加线粒体的产能,以此来改善和纠正心衰。 Objective The purpose of this study was to observe the effect of Yiqi Huoxue Recipe on myocardial energy metabolism in rats with heart failure(HF)after myocardial infarction.Methods Eighty SPF male Sprague-Dawley rats were selected,and 20 of them were randomly selected as the blank control group.The remaining 60 rats were treated with left anterior descending coronary artery ligation combined with exhaustive swimming and diet to make heart failure.The rats with successful modeling were randomly divided into western medicine group,model group and Yiqi Huoxue Recipe group.The blank control group and the model group were given the same volume of distilled water once a day.The western medicine group was given lisinopril 1.5 mg/(kg·d)once a day,and the Yiqi Huoxue Recipe group was given 10.0 g/(kg·d)once a day.After continuous administration for 28 days,the rats were fasted for 12 hours,and then the myocardial tissue and plasma were taken out.ELISA method was used to detect BNP concentration.The immunoblotting and real-time fluorescent quantitative PCR detected PGC-1αgene and protein expression.Results Compared with those of the blank control group,the expressions of PGC-1αmRNA and protein in the Yiqi Huoxue Recipe group,the model group and the western medicine group decreased,and the BNP concentration increased,which was statistically significant(P<0.05).Compared with those of the model group,the expression of PGC-1αmRNA and protein in the western medicine group and Yiqi Huoxue Recipe group increased,and the BNP concentration decreased,which was statistically significant(P<0.05).Both the western medicine group and the Yiqi Huoxue Recipe group had no statistical difference(P>0.05).Conclusion Yiqi Huoxue Recipe can improve and correct heart failure by activating PGC-1α,increasing myocardial energy metabolism and increasing mitochondrial productivity.
