芪蓟肾康汤对血管紧张素Ⅱ诱导的人肾小球系膜细胞FN、TGF-β1影响的研究 被引量:1

摘要 目的观察芪蓟肾康汤对AngⅡ诱导人肾小球系膜细胞FN及TGF-β1表达,探讨不同剂量芪蓟肾康汤改善肾小球硬化作用机制。方法用酶联免疫法吸附试验(ELISA)法检测空白组,模型组,替米沙坦组及低、中、高剂量组芪蓟肾康汤含药血清培养AngⅡ诱导人肾小球系膜细胞株FN、TGF-β1的表达。结果24 h和48 h的FN、TGF-β1含量表达,模型组高于正常组(明显)(P<0.01);低、中、高剂量组芪蓟肾康汤较模型组减少,作用时间越长抑制效果越好(P<0.01);中、高剂量组芪蓟肾康汤低于西药组,中药组间剂量芪蓟肾康汤越高抑制效果越好(P<0.05)。结论芪蓟肾康汤可有效抑制FN、TGF-β1的表达,从而抑制AngⅡ诱导人肾小球系膜细胞增殖,潜在改善肾小球硬化程度。 Objective To observe the effect of Qiji Shenkang Decoction on expressions of FN and TGF-β1 of human glomerular mesangial cells induced by AngⅡand explore the mechanism of Qiji Shenkang Decoction in the prevention and treatment of renal glomerular sclerosis.Methods ELISA method was used to detectthe expressions of FN and TGF-β1of human glomerular mesangial cell lines induced by AngⅡin the blank group,model group,the western medicine group(telmisartan group)and the low,middle and high dose ofserum-containing Qiji Shenkang Decoction groups.Results At 24 h and 48 h,the contents of FN and TGF-β1were significantly higher in the model group than those in the normal group(P<0.01).Compared with the model group,the effect of Qiji Shenkang Decoction low,middle and high dose groups was significantly reduced,and the effect was more obvious with longer time(P<0.01).The middle and high dose groups’effective rate was less than that of the western medicine group.Among the three groups of Qiji Shenkang Decoction,the higher dose had a better inhibition effect(P<0.05).Conclusion Qiji Shenkang Decoction can effectively inhibit the expressions of FN and TGF-β1,thereby inhibiting the proliferation of human mesangial cells and potentially delaying the process of glomerular sclerosis.
