铁-邻菲罗啉体系共振散射法测定抗坏血酸 被引量:4

摘要 研究了抗坏血酸(Vc)还原铁(Ⅲ)-邻菲罗啉(Phen)体系的光谱特性,考察了各种影响因素,确定了体系的共振散射强度与Vc浓度之间的关系,建立了测定Vc含量的新方法.结果表明,在pH6.5的B-R(Britton-Robinson)缓冲溶液中,Vc作为还原剂,将Fe(Ⅲ)还原为Fe(Ⅱ),Fe(Ⅱ)继而与邻菲罗啉配位形成[Fe(phen)3]2+配合物,其在384nm波长处产生1个较强的共振散射峰.并且随着抗坏血酸用量的增加,体系的共振散射强度随之降低.在最优实验条件下,在0.040~6.0mg/L范围内,体系的共振散射强度与抗坏血酸浓度之间呈良好的线性关系.该方法简便快速、灵敏度高、选择性好,可直接用于药物等实际样品中抗坏血酸含量的测定,结果较满意. The spectral characteristic of Fe(Ⅲ)-Phen reduced by ascorbic acid was studied,and the various influencing factors were examined,with the relationship between the resonance scattering intensity and Vc concentration determined,thus a new method for the determination of Vc content was established.It is found that in the pH 6.5 B-R(Britton Robinson)buffer solution,ascorbic acid(Vc)used as the reduction agent,after Fe(Ⅲ)is reduced to Fe(Ⅱ),and then Fe(Ⅱ)coordinates with phenanthroline(phen)to form[Fe(phen)3]2+complex,which can produce a strong resonance scattering peak at the wavelength of 384 nm.And with the increase of dosage of ascorbic acid,the resonance scattering intensity decreases.Under the optimum experimental conditions,there is a good linear relationship between the system resonance scattering intensity and ascorbic acid concentration in the range of 0.040~6.0 mg/L.The method is convenient,of high sensitivity and good selectivity,thus can be directly used for the determination of ascorbic acid content in the actual sample of drugs with a satisfactory result.
