摘要
为研究红茂草多糖提取工艺及其对免疫低下小鼠免疫功能的影响,以红茂草多糖含量为评价指标,在前期试验的基础上进行正交试验设计,确定红茂草多糖的最佳提取工艺。提取的不同剂量红茂草多糖对BALB/c小鼠进行灌胃,之后以环磷酰胺为免疫抑制剂,构建小鼠免疫低下模型,综合评价红茂草多糖对免疫低下小鼠免疫功能的调节作用。结果表明,红茂草多糖的最佳提取工艺为超声波处理时间35 min、料液比1∶20(g∶mL)、索氏回流时间160 min、醇沉28 h、pH值为5和体积分数80%的乙醇,其产率可达1.19%;红茂草多糖能显著提高免疫低下小鼠的免疫功能,对免疫失衡机体具有一定的调节作用。红茂草具有良好的开发利用前景。
In order to investigate the extraction technology of polysaccharides from Dicranostigma leptodum and their effects on the immune function of immunocompromised mice,using D.leptodum polysaccharides content as evaluation index,orthogonal experiments were determined on the basis of former experiments,and the optimum extraction technology of D.leptodum polysaccharides was determined.The BALB/c mice were given different doses of extracted polysaccharides by gavage,and then cyclophosphamide was used as an immunosuppressant to construct a mouse immunosuppressant model,and the regulatory effect of D.leptodum polysaccharides on immune function of immunocompromised mice was comprehensively evaluated.The results showed that the optimal extraction technology of D.leptodum polysaccharides was as follows:ultrasonic treatment time 35 min,solid-liquid ratio 1∶20(g∶ml),Soxhlet reflux time 160 min,pH 5,ethanol volume fraction 80%,and precipitation 28 h,and the yield of polysaccharides was 1.19%.D.leptodum polysaccharides could significantly improve the immune function of immunocompromised mice,and had a certain regulation effect on immune imbalance,which had a good prospect of development and utilization.
