摘要
目的分离鉴定小花黄堇中的特征性成分奥柯紫堇明碱,并测定其含量。方法用色谱和波谱技术从小花黄堇中分离得到奥柯紫堇明碱,采用HPLC法测定小花黄堇中的奥柯紫堇明碱,用Thermo BDS HYPERSIL C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.2%冰乙酸溶液(三乙胺调p H5.0)为30∶70,流速1 m L·min^-1,检测波长290 nm,柱温30℃。结果33.28~665.60 ng奥柯紫堇明碱与峰面积的线性关系良好(r2=0.9999),平均回收率为98.23%,RSD=3.86%。结论分离得到的奥柯紫堇明碱纯度高,可作为含量测定用对照品;所用定量方法简便、准确、重复性好,可为小花黄堇的质量控制提供参考。
OBJECTIVE To separate and identify chemical marker ochotensimine in the Corydalis racemosa(Thunb.)Pers.To establish an HPLC method for determination the content of ochotensimine in C.racemosa.METHODS Ochotensimine was separated and identified from C.racemosa by chromatographic technique and spectroscopic method for the first time.The content of ochotensimine was determined by HPLC analysis on an Thermo BDS HYPERSIL C18 column(250 mm×4.6 mm,5μm)with acetonitrile-0.2%glacial acetic acid solution(contain triethylamine for p H5.0)30∶70 solution as mobile phase with flow rate of 1 m L·min^-1.The detection wavelength was set at 290 nm and the column temperature was 30℃.RESULTS The HPLC method showed good linearity within the range of 33.28-665.60 ng for ochotensimine(r2=0.9999).The average recovery was98.23%,and the RSD was 3.86%.CONCLUSION Ochotensimine obtained by this separation method is of high purity,and it can be adopted as the reference substance.The established quantitative method is simple,accurate and reproducible,and can be used for the quality control of C.racemosa.
