摘要
目的研究贝母素乙(PMI)联合奥沙利铂对胃癌MKN45细胞增殖和凋亡作用。方法CCK8法检测PMI联用奥沙利铂处理24、48、72 h后胃癌MKN45细胞的OD值计算细胞增殖率以及凋亡率,筛选最佳作用浓度及作用时间;结晶紫染色法观察PMI联用奥沙利铂对胃癌MKN45细胞增殖的影响;Western blot检测PMI联用奥沙利铂处理后对细胞凋亡相关蛋白的表达。结果PMI在浓度范围10~20μg/mL对胃癌MKN45细胞无抑制作用,与奥沙利铂组比较,PMI联用奥沙利铂组干预胃癌MKN45细胞48 h和72 h后抑制率升高,IC50降低,差异有统计学意义(P<0.05)。PMI联用奥沙利铂可抑制胃癌MKN45细胞克隆,下调胃癌MKN45细胞Caspase 3蛋白表达量,增加Cleaved-parp和Cleaved-caspase 3蛋白表达量。结论奥沙利铂在体外实验抑制胃癌MKN45细胞增长,PMI联用奥沙利铂可产生协同作用。
Objective The effect of peiminine(PMI)combined with oxaliplatin on the proliferation and apoptosis of gastric cancer MKN45 cells.Methods The proliferation and apoptosis rate of gastric cancer MKN45 cells treated with PMI combined with oxaliplatin for 24,48 and 72 h,were detected by CCK8.The effect of PMI and oxaliplatin combination treatment on cell proliferation of MKN45 was detected by crystal violet staining.The expression of apoptosis-related proteins in the MKN45 treated by PMI combined with oxaliplatin was detected by Western blot.Results PMI had no inhibitory effect on MKN45 cell in the concentration range of 10-20μg/mL.Compared with oxaliplatin group,the inhibition rate of MKN45 cells in PMI combined with oxaliplatin group was increased and its IC50 was decreased after 48 h and 72 h treatment,and the difference was statistically significant(P<0.05).In the PMI combined with oxaliplatin group,the cell clone number of MKN45 cells was reduced,the expression of Caspase-3 protein was down-regulated,while the protein expression of Cleaved-parp and Cleaved-caspase-3 was increased.Conclusion Oxaliplatin can inhibit the growth of human gastric cancer cell line MKN45 in vitro,and PMI combined with oxaliplatin has obvious synergistic effect.
