肉苁蓉苯乙醇总苷对庆大霉素诱导急性肾损伤大鼠的药效学研究 被引量:11

摘要 目的研究肉苁蓉苯乙醇总苷(phGs)对庆大霉素诱导大鼠急性肾损伤的保护作用。方法将54只雄性SD大鼠随机分为10 d、15 d空白对照组(生理盐水10 mL/kg体质量),10 d、15 d模型组(生理盐水10 mL/kg体质量),10 d、15 d phGs高剂量组(phGs 500 mg/kg体质量),10 d、15 d phGs低剂量组(phGs 250 mg/kg体质量)以及21 d模拟临床治疗组(生理盐水10 mL/kg体质量),每组6只,除10 d、15 d空白对照组外其他各组大鼠连续10 d腹腔注射庆大霉素(75 mg/kg体质量)建立急性肾损伤模型。各组实验结束后留取大鼠24 h尿液并记录尿量,腹主动脉取血,取双侧肾脏并计算肾脏指数。采用全自动生化分析仪测定各组大鼠血清中肌酐(Scr)和尿素氮(BUN)含量,采用生化试剂盒检测各组大鼠肾脏组织中丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)表达量。结果与空白对照组比较,10 d、15 d模型组大鼠血清Scr、BUN、24 h尿量及肾脏组织中MDA表达量均升高,肾脏组织中SOD、CAT、GSH-PX酶的活性均降低,差异有统计学意义(P<0.01)。与模型组比较,10 d、15 d phGs高、低剂量组大鼠血清Scr、BUN、24 h尿量及肾脏组织中MDA表达量均降低,肾脏组织中SOD、CAT、GSH-PX酶的活性均升高,差异有统计学意义(P<0.05)。结论肉苁蓉苯乙醇总苷对庆大霉素诱导的大鼠急性肾损伤具有一定保护作用。 Objective To study the protective effect of phenylethanoid glycosides(phGs)on gentamicin-induced acute kidney injury in rats.Methods A total of 54 male SD rats were randomly divided into the control group(10 d,15 d,normal saline 10 mL/kg),model group(10 d,15 d,normal saline 10 mL/kg),phGs 500 mg/kg group(10 d,15 d),phGs 250 mg/kg group(10 d,15 d),and simulated clinical group(21 d,normal saline 10 mL/kg),each group hadsix rats.The rat model of acute kidney injury was established by intraperitoneal injection of gentamicin at 75 mg/kg for 10 days.At the end of experiment,the 24 h urine of ratswascollected,bloodof rats was taken from the abdominal aorta,bilateral kidneysof rats were taken and the renal index was calculated.Serum creatinine(Scr),blood urea nitrogen(BUN)were measuredby auto-analyzer.Superoxide dismutase(SOD),glutathione peroxidase(GSH-PX),catalase(CAT)and malonaldehyde(MDA)levels in renal tissue were detected.Results Compared with the blank control group,the Scr,BUN,24 h urine volume and MDA expression level in kidney tissue were increased in the 10 d and 15 d model group,and the activities of SOD,CAT and GSH-PX in renal tissue were decreased significantly(P<0.01).Compared with the model group,the serum Scr,BUN,24 h urine volume and the expression of MDA in kidney tissue were decreased in 10 d and 15 d phGs high and low dose groups(P<0.05),while the activities of SOD,CAT and GSH-PX in kidney tissue were increased(P<0.05).Conclusion phGs had a protective effect on gentamicin-induced acute kidney injury in rats.
